摘  要:目的:探讨姜黄素对胰腺癌干细胞自我更新及其Wnt/β-Catenin信号通路活性的影响。方法:(1)通过细胞球培养法获取Panc1胰腺癌干细胞。(2)通过MTT法检测姜黄素对Panc1胰腺癌干细胞自我更新能力的影响。通过实时定量PCR检测姜黄素对Panc1胰腺癌干细胞中Wnt1、β-Catenin、Cyclin D1 mRNA表达水平的影响。通过细胞免疫组化检测姜黄素对Panc1胰腺癌干细胞中β-Catenin蛋白表达的影响。结果:(1)Panc1胰腺癌细胞悬浮培养时全部细胞聚集成球状。(2)悬浮培养5天后得到的Panc1胰腺癌干细胞体外培养时,加入20μM姜黄素后,细胞增殖能力明显受到抑制(P<0.01);细胞中Wnt1、β-Catenin、Cyclin D1的mRNA转录水平明显下调,β-Catenin蛋白表达也明显下调,细胞质、细胞核均未见β-Catenin表达。结论:姜黄素可显著抑制胰腺癌干细胞自我更新,并可显著抑制其Wnt/β-Catenin信号通路活性,抑制该通路活性可能是姜黄素抑制胰腺癌干细胞自我更新的机制之一。Objective:To investigate the effect of curcumin on the self-renewal and activity of Wnt/β-catenin signal pathway of pancreatic cancer stem cells(CSCs).Methods:(1)Pnac1 pancreatic CSCs were obtained from cell spheres culture.(2)The effect of curcumin on self-renewal ability of Panc1 pancreatic CSCs were detected by MTT assay.The effect of curcumin on Wnt1,β-catenin and Cyclin D1 mRNA level in Panc1 pancreatic CSCs was detected by real-time quantitative PCR.The effect of curcumin on the expression of β-catenin protein in Panc1 pancreatic CSCs was detected by cell immunohistochemistry.Results:(1)All the Panc1 pancreatic cancer cells aggregated into cell spheres in suspensionculture.(2)Panc1 pancreatic CSCs obtained by 5 days spheres culture were cultered in vitro,after 20μM curcumin was added,Panc1 pancreatic CSCs’s proliferation was significantly inhibited(P<0.01),the mRNA transcription levels of Wnt1,β-catenin and Cyclin D1 were significantly down-regulated and the expression levels ofβ-catenin protein were significantly suppressed,no β-catenin was showed in the cytoplasm or nucleus of the cells.Conclusion:Curcumin can inhibit the self-renewal and the activity of Wnt/β-catenin signal pathway of pancreatic CSCs and the suppression of Wnt/β-catenin signaling pathway may be one of the mechanisms of curcumin inhibiting the self-renewal ability in pancreatic CSCs.
