摘  要:目的通过敲低长链非编码RNA AC007392.4的表达,探讨其对结直肠癌细胞增殖、凋亡及周期的影响及潜在分子机制。方法通过癌症基因组图谱(the Cancer Genome Atlas,TCGA)分析AC007392.4在结直肠组织的表达情况;通过皮尔森相关系数构建AC007392.4基因共表达网络,并对共表达基因进行富集分析;通过脂质体转染法敲低结直肠癌细胞系SW480中AC007392.4的表达,CCK-8法检测AC007392.4对结直肠癌细胞增殖的影响;流式细胞仪检测AC007392.4对细胞凋亡及细胞周期的影响;Real-time PCR方法检测敲低AC007392.4对下游通路关键分子mRNA的影响。结果 AC007392.4在结直肠癌中表达上调(P=0.041),且与病理分期有关(P=0.044);共有2354个蛋白编码基因与AC007392.4表达具有高度相关性,且这些蛋白编码基因显著地富集在DNA损伤修复与细胞周期相关的通路中;敲低AC007392.4抑制结直肠癌细胞增殖、促进其凋亡;敲低AC007392.4后可使细胞周期G2/M期发生阻滞;敲低AC007392.4后,CDK1的mRNA表达水平明显降低(P<0.05)。结论 AC007392.4在结直肠癌中表达量上调,敲低后抑制结直肠癌细胞的增殖并促进凋亡;AC007392.4可能通过调控细胞周期参与了结直肠癌的发生发展。Objective To investigate the effect of AC007392.4 on the proliferation,apoptosis and cycle of colorectal cancer cell and its potential molecular mechanism by knocking down the expression of long non-coding RNA AC007392.4.Methods The expression of AC007392.4 in colorectal tissues was analyzed by the Cancer Genome Atlas(TCGA),the AC007392.4 gene co-expression network was constructed by Pearson correlation coefficient,and enrichment analysis on co-expressed genes was performed.The AC007392.4 expression in colorectal cancer cell line SW480 was downregulated by liposome transfection.CCK-8 method was used to detect the effect of AC007392.4 on the proliferation of colorectal cancer cells.Flow cytometry was used to detect the effect of AC007392.4 on cell apoptosis and cell cycle.Real-time PCR was used to detect the effect of knockdown of AC007392.4 on the mRNA of key molecules in downstream pathways.Results AC007392.4 was up-regulated in colorectal cancer(P=0.041),and was related to pathological staging(P=0.044).A total of 2354 protein-coding genes were highly correlated with the expression of AC007392.4,and these protein-coding genes were significantly enriched in pathways related to DNA damage repair and cell cycle.Knockdown of AC007392.4 inhibited the proliferation of colorectal cancer cell and promotes their apoptosis.Knockdown of AC007392.4 could also block the G2/M phase of the cell cycle.After knocking down AC007392.4,the mRNA expression level of CDK1 was significantly reduced(P<0.05).Conclusion The expression of AC007392.4 in colorectal tissues was analyzed by the Cancer Genome Atlas(TCGA),the AC007392.4 gene co-expression network was constructed by Pearson correlation coefficient,and enrichment analysis on co-expressed genes was performed.The AC007392.4 expression in colorectal cancer cell line SW480 was downregulated by liposome transfection.CCK-8 method was used to detect the effect of AC007392.4 on the proliferation of colorectal cancer cells.Flow cytometry was used to detect the effect of AC007392.4 on cell
