摘　　要：目的分析环状RNA001569(circ_001569)在胆囊癌(Gallbladder carcinoma,GBC)组织中的表达及其与患者临床特征、预后的关系。方法回顾性收集2013年1月至2018年12月间于我院行根治性手术的62例GBC患者的肿瘤样本及临床资料,并定期随访。选择同期入院的20例胆囊腺瘤及20例慢性胆囊炎患者作为对照组。qRT-PCR检测患者circ_001569的表达情况,分析其与GBC患者临床病理特征的关系。Keplan-Meier生存曲线、log-rank检验及Cox回归分析探讨circ_001569对GBC患者总体生存率(Overall survival,OS)的影响。结果 GBC组织中circ_001569的表达水平显著高于胆囊炎和胆囊腺瘤,且差异具有统计学意义(P<0.001)。不同肿瘤分化程度、不同TNM分期的患者circ_001569表达有显著差异。Keplan-Meier生存曲线提示,circ_001569表达水平较高的患者有较差的OS。Cox多因素分析提示,circ_001569是GBC的独立预后因素。结论 Circ_001569在GBC组织中高表达,与患者肿瘤分化程度、TNM分期密切相关,并影响患者的预后。Objective To analyze the expression of circular RNA 001569(circ001569)in gallbladder carcinoma(GBC)and its relation to clinicopathological features and prognosis.Methods The tumor samples and clinical data of 62 patients with GBC who underwent radical surgery in our hospital from January 2013 to December 2018 were collected and regularly followed up.20 patients with gallbladder adenoma and 20 patients with chronic cholecystitis were selected as control groups.qRT-PCR was used to detect the expression of circ001569.The relationship between the expression of circ001569 and the clinicopathological features of patients with GBC was analyzed.Keplan-Meier survival curve,log-rank test and Cox regression analysis were used to investigate the effect of circ001569 on the overall survival(OS).Results The expression level of circ001569 in GBC was significantly higher than that in cholecystitis and gallbladder adenoma,and the difference was statistically significant(P<0.001).The expression of circ001569 was significantly associated with tumor differentiation and TNM stage.Keplan-Meier survival curve indicated that patients with higher level of circ001569 had worse OS.Cox multivariate analysis showed that circ001569 was an independent prognostic factor for GBC.Conclusion Circ001569 is highly expressed in GBC,which is closely related to tumor differentiation and TNM stage,and affects the prognosis of patients.
