儿童激素性高眼压与外周血淋巴细胞糖皮质激素受体表达相关性研究

摘　　要：目的:检测激素性高眼压患儿外周血淋巴细胞糖皮质激素受体(GR),GRα和GRβ的表达,研究其与儿童激素性高眼压的相关性。方法:收集20例激素性高眼压患儿(高眼压组),20例使用激素后眼压正常患儿(对照组),20例健康儿童(健康组)的外周血,提取淋巴细胞RNA,采用半定量逆转录-聚合酶链反应(RT-PCR)法检测淋巴细胞内GRα、GRβmRNA表达水平。结果:儿童外周血淋巴细胞有GRα、GRβmRNA表达,以GRα为主。高眼压组的GRαmRNA半定量比值为1.29±0.14,与对照组1.23±0.13及健康组1.28±0.08相比,3组之间的表达差异无显著性意义(P>0.05)。高眼压组的GRβmRNA半定量比值为1.24±0.04,与对照组1.22±0.09及健康组1.19±0.07相比,3组之间的表达差异无显著性意义(P>0.05)。结论:未发现儿童外周血淋巴细胞糖皮质激素受体GRα和GRβ的表达水平与激素性高眼压之间相关性。Objective:To investigate the relationship between glucocorticoid ocular hypertension and expression of glucocorticoid receptor m RNA(GR)in peripheral blood lymphocyte in children. Methods:Peripheral blood from 20 children with glucocorticoid ocular hypertension(ocular hypertension group),20 children without glucocorticoid ocular hypertension after glucocorticoid treat(control group),and 20 healthy children(healthy group)was collected to detect the expression level of GRαand GRβ mRNA in peripheral blood lymphocyte by semi-quantitative reverse transcriptional polymerase chain reactin(RT-PCR). Results:There were both GRα and GRβ mRNA in peripheral blood lymphocyte, and the content of GRα is higher than GRβ. GRα/actin in ocular hypertension group was1.29 ± 0.14,which has no significance difference among control group 1.23±0.13 and healthy group 1.28±0.08. GRβ/actin in ocular hypertension group was 1.24 ±0.04, Compared with the control group 1.22±0.09 and the healthy group 1.19±0.07, there was no significant difference in the expression of GRβbetween the three groups(P > 0.05).Conclusion:No correlation was found between the expression of glucocorticoid receptor GRα and GRβ in peripheral blood lymphocytes of children and glucocorticoid ocular hypertension.
