摘  要:目的:建立表达人癌胚抗原(CEA)的小鼠肝癌细胞模型。方法:用脂质体介导转染法将含人CEA-cDNA的逆转录病毒重组质粒pLXSN-CEA导入小鼠肝癌细胞,利用含G418的选择培养基筛选及放免法测定癌细胞CEA表达量进行鉴定。结果:成功建立表达人CEA的小鼠肝癌细胞模型。结论:利用基因转染技术能将人癌胚抗原cDNA基因导入小鼠肝癌细胞中,并能在选择培养基中生长,且表达人的癌胚抗原。Objective:To establish murine Hepa cancer cell model expressing human carcinoembryonic antigen (CEA).Methods: The recombinant plasmid pLXSN-CEA with human CEAswhd pcoN - CEA with human CEA-cDNA was transferred into the ng lipofecedne. FuIther screening with G4l8 and edng of CEA expressed by the Hepa cancer cells with radio-immunology assay were taken to identify the CEA positive caner cells.Results: The Hepa caner cells model of murine expressing human CEA was obtained successfully. Conclusion:Human CEA - cDNA plasmid could be transferred into murine Hepa cancer cells by DNA transfer techniques.
