摘 要:目的采用生物信息学方法分析miR-486-3p靶基因在甲状腺乳头状癌(PTC)中的差异表达情况及临床意义。方法根据肿瘤基因图谱中PTC肿瘤样本及配对样本的表达数据筛选miR-486-3p差异表达基因,使用ClusterProfiler R包对差异表达基因进行GO和KEGG注释,ggplot2 R包绘制GO二级分类柱形图及KEGG注释统计图,将预测miR-486-3p靶基因并与差异表达基因取交集,采用Survival R包对样本进行生存分析,绘制Kaplan-Meier生存曲线。结果共筛选出99个差异表达基因,其中74个基因上调,25个基因下调,差异表达的基因多与细胞组织结构、形态发育、ERK1/2通路等功能调控相关,主要涉及病毒感染、细胞周期、泛素化、蛋白水解、p53信号通路、细胞衰老等通路。miR-486-3p低表达的PTC患者总生存率低于高表达的患者(P<0.05)。结论miR-486-3p可在PTC中参与调节肿瘤细胞的生物学功能,且低表达miR-486-3p的PTC患者预后不良,提示miR-486-3p或作为抑癌基因及生物标志物在PTC发生、发展中起重要作用。Objective To analyze the relationship between miR-486-3p expression and thyroid papillary carcinoma(PTC)with bioinformatics.Methods According to the expression profile of PTC tumor samples in The Cancer Genome Atlas(TCGA)detabase,differential expression genes were screened.Clusterprofiler R package was used to annotate differential expression genes with GO and KEGG,and ggplot2 R package was used to draw the GO secondary classification column and KEGG annotation statistical chart.miR-486-3p target genes were predicted and R package was used for survival analysis and the Kaplan Meier survival curve was generated.Results A total of 99 differentially expressed genes were screened out,among which 74 genes were up-regulated and 25 genes were down-regulated.The differentially expressed genes were mostly related to cell structure,morphological development,ERK1/2 pathway and other functions,mainly involving virus infection,cell cycle,ubiquitination,proteolysis,p53 signal pathway,cell aging and other pathways.The overall survival rate of PTC patients with down-regulated mir-486-3p expression was low(P<0.05).Conclusion The miR-486-3p may regulate the biological functions of tumor cells in PTC,and the prognosis of PTC patients with low expression of miR-486-3p is poor,which suggested the miR-486-3p may play an important role in the development of PTC as a tumor hall marker gene.
