摘　　要：目的:探讨过表达原钙黏蛋白10(PCDH10)基因对结直肠癌细胞增殖和迁移的影响及机制。方法:采用qRT-PCR检测不同结直肠癌细胞(SW620、HT29、LoVo、HCT116)及正常结肠上皮细胞NCM460中PCDH10 mRNA的表达水平。以含pcDNA-PCDH10过表达载体质粒的慢病毒感染结直肠癌SW620细胞(PCDH10组),以感染含空载质粒的慢病毒为阴性对照,未感染细胞为空白对照。采用qRT-PCR和Western blot法检测上述3组细胞的转染效率,MTT法和克隆形成实验检测细胞增殖能力,Transwell实验检测细胞迁移能力,采用Western blot法检测细胞中增殖细胞核抗原(PCNA)和基质金属蛋白酶-2(MMP-2)的表达水平。结果:PCDH10在结直肠癌细胞中的表达水平均低于正常结肠上皮细胞NCM460(P<0.05)。与二对照组相比,感染组SW620细胞中PCDH10 mRNA和蛋白表达水平升高(P<0.05),细胞增殖和迁移受抑,同时细胞中PCNA和MMP-2蛋白的表达水平降低(P<0.05)。结论:PCDH10基因在结直肠癌细胞中呈低表达,过表达PCDH10能够抑制结直肠癌SW620细胞增殖和迁移能力,该过程可能与抑制PCNA和MMP-2蛋白的表达有关。Aim:To investigate the effects and mechanism of protocadherin 10(PCDH10)gene overexpression on proliferation and migration of colorectal cancer cells.Methods:qRT-PCR was used to detect the expression of PCDH10 mRNA in different colorectal cancer cell lines(SW620,HT29,LoVo,HCT116)and normal colonic epithelial cell line NCM460.SW620 cells were infected by pcDNA-PCDH10 overexpression vector lentivirus,those infected by blank plasmid vector lentivirus were used as negative control,and those not infected were blank control.qRT-PCR and Western blot were used to detect the transfection efficiency.MTT and clone formation assays were used to detect cell proliferation.Transwell assay was used to detect cell migration.Western blot was used to detect the expressions of proliferating cell nuclear antigen(PCNA)and matrix metalloproteinase-2(MMP-2).Results:The expression level of PCDH10 in colorectal cancer cells was significantly lower than that in normal colonic epithelial NCM460 cells(P<0.05).PCDH10 overexpression could up-regulate the expressions of PCDH10 mRNA and protein in SW620 cells(P<0.05).Overexpression of PCDH10 could inhibit cell proliferation and migration,and down-regulate the expressions of PCNA and MMP-2 in SW620 cells(P<0.05).Conclusion:PCDH10 gene is down-regulated in colorectal cancer cells.Overexpression of PCDH10 can inhibit the proliferation and migration of SW620 cells,which may be related to the inhibition the expressions of PCNA and MMP-2 protein.
