摘 要:目的:探索神经干细胞对大鼠抑郁症模型的干预效果及对海马神经元修复的影响。方法:采用慢性不可预见性应激抑郁症模型(CUMS)方法建立大鼠抑郁症模型;神经干细胞分离、培养和鉴定;定位海马区域注射神经干细胞治疗抑郁症大鼠,观察相关指标变化。结果:体重及行为学指标(糖水偏好、旷场实验)提示注射神经干细胞可明显改善大鼠抑郁样行为(P<0.05),病理学结果提示神经干细胞可改善抑郁症大鼠海马细胞受损情况,RT-PCR结果提示神经干细胞可提高抑郁症大鼠海马区神经相关因子的表达。结论:海马区注射神经干细胞可改善大鼠抑郁样症状,其作用机制可能与修复海马神经元有关。Objective:To evaluate the effect of neural stem cells(NSC)on the depression model of rats and the effect on the repair of hippocampal neurons.Methods:1.To establish rats’CUMS depression model;2.To isolate,culture and identify neural stem cells;3.To treat depression rats by injecting NSC into hippocampus.Results:Weight and behavioral indexes(sugar water preference,open-field experiment)suggested that NSC could significantly improve depression like behavior(P<0.05).Pathological results showed that NSC could repair hippocampal neural cells which were damaged in depression rats.RT-PCR results indicated that NSC could increase the expression of neural related cytokine factors in the hippocampal region of depression rats.Conclusion:Injection of NSC into hippocampus can improve depression like symptoms in rats,and its mechanism may be to repair hippocampal neurons.
