摘　　要：目的:探讨芳香烃受体(aryl hydrocarbon receptor,AhR)基因表达与弥漫大B细胞淋巴瘤(Diffuse large B cell lymphoma,DLBCL)患者临床病理特征及预后的关系。方法:利用GTEx、TCGA及GEO数据集分析正常人及DLBCL患者AhR mRNA水平,免疫组化检测组织芯片AhR蛋白水平,分析AhR基因表达与DLBCL患者临床病理特征相关性及患者生存预后相关性,GSEA法预测AhR调控基因及潜在相关通路。结果:AhR在DLBCL中高表达(P<0.05)。AhR mRNA水平在非生发中心型(non-germinal center B-cell-like lymphoma,non-GCB)淋巴瘤患者中显著高于生发中心型(germinal center B-cell-like lymphoma,GCB)患者(P<0.05),AhR mRNA表达水平与DLBCL患者其他指标无显著相关性,包括远期生存率,AhR不是DLBCL患者的独立预后因素。高表达AhR组富集于TCA循环、过氧化氢酶体,以及缬氨酸、亮氨酸和异亮氨酸降解,低表达组富集于氧化磷酸化等代谢信号通路。结论:AhR高表达与non-GCB显著相关而与其他临床相关病理指标及预后无显著相关,AhR可能在DLBCL发生过程中发挥作用,与淋巴细胞物质和能量代谢有关。Objective To investigate the association between AhR(aryl hydrocarbon receptor)gene expression and clinicopathological features and prognosis of patients with large B cell lymphoma(DLBCL).Methods The AhR mRNA levels in healthy people and DLBCL patients were analyzed using datasets from GTEX,TCGA and GEO.The AhR protein levels in tissue array were determined using immunohistochemistry.The correlation between AhR gene expression and clinical pathological characteristics and prognosis of DLBCL patients was analyzed.The regulatory factors and potential related pathways of AhR were predicted using GSEA.Results AhR was highly expressed in DLBCL(P<0.05).The level of AhR mRNA in non-GCB(non-germinal center B-cell-like lymphoma)lymphoma patients was significantly higher than that in GCB(germinal center B-cell-like lymphoma)group(P<0.05).AhR expression had no significant correlation with other clinical pathological indicators of DLBCL patients,including the long-term survival rate.AhR was not an independent prognostic factor in DLBCL patients.TCA cycle,catalase and degradation of valine,leucine and isoleucine were enriched in the high expression AhR group,and oxidative phosphorylation and other metabolic signaling pathways were enriched in the low expression AhR group.Conclusion The high expression of AhR was significantly correlated with non GCB,but not with other clinicopathological parameters and prognosis of DLBCL.AhR may play a role in the development of DLBCL and be involved in regulation of substance and energy metabolism in lymphocytes.
