摘 要:目的探讨MDM2抑制剂MI-219对前列腺癌细胞株的抑制作用及其机制。方法通过辐照和MI-219作用后检测细胞克隆形成率,利用流式细胞术检测空白组、辐照组、MI-219组以及辐照+MI-219组细胞周期。分别对动物体内肿瘤进行辐照和MI-219处理,检测细胞系以及肿瘤组织中P21、BAX的表达。结果22RV1、LNCa P、PC3和DU145克隆形成率明显低于对照组,流式细胞分析显示细胞系经MI-219+辐照作用后增殖阻滞。动物实验中,MI-219+辐照可以显著抑制肿瘤的生长。PCR检测表明细胞系和肿瘤组织中P21、BAX的mRNA表达明显升高,并且辐照+MI-219组mRNA表达明显高于辐照组、MI-219组。结论MDM2抑制剂MI-219对前列腺癌细胞产生增敏作用,导致细胞更容易凋亡,这为前列腺癌的治疗提供新思路,有望提高前列腺癌患者的生存率。Objective To investigate the inhibitory effect and mechanism of MDM2 inhibitor MI-219 on prostate cancer cell lines.Methods The cell clone formation rate was detected after radiation and MI-219,and cell cycle of blank group,irradiation group,MI-219 group and radiation+MI-219 group was detected by flow cytometry.The expressions of P21 and BAX in cell lines and tumor tissues were detected by radiation and MI-219 treatment,respectively.Results The clone formation rates of 22 RV1,LNCa P,PC3 and DU145 were significantly lower than those of the control group.Flow cytometry analysis showed that the cell lines were inhibited after MI-219+radiation.In animal experiments,MI-219+radiation can significantly inhibit tumor growth.PCR detection showed that the mRNA expressions of P21 and BAX in cell lines and tumor tissues were significantly increased,and the mRNA expressions of group radiation+MI-219 were significantly higher than those of group radiation and MI-219.Conclusion MDM2 inhibitor MI-219 has a sensitizing effect on prostate cancer cells,which leads to cell apoptosis.This provides new ideas for the treatment of prostate cancer and is expected to improve the survival rate of patients with prostate cancer.
