摘 要:(1)目的研究具有不同晶面能的纳米硫化镉(nano-CdS)对小鼠血液学指标及肝脏组织损伤作用的差异。(2)方法选取昆明种小鼠24只,随机分为三组,分别为对照组、CdS1组和CdS2组,每组8只。采用腹腔注射法染毒,CdS1组和CdS2组分别注射0.2mL的nano-CdS1和nano-CdS2混悬液(20mg/kg),对照组注射0.2mL的生理盐水,每周1次,连续4周。染毒结束后检测小鼠血液学指标变化及血清谷草转氨酶(AST)和谷丙转氨酶(ALT)含量,并计算比值;用HE染色法观察小鼠肝脏组织病理学变化;检测小鼠血清中及肝组织内丙二醛(MDA)、一氧化氮(NO)含量。(3)结果与对照组比较,CdS2组小鼠血液中血红蛋白含量、红细胞压积及红细胞平均体积等指标均降低,而白细胞数、血小板含量及AST/ALT比值升高,差异有统计学意义(P<0.05)。CdS1组和CdS2组肝脏组织均有不同程度病理损伤,且CdS2组损伤较明显。CdS2组血清中MDA、NO含量较CdS1组显著升高,差异有统计学(P<0.05);肝脏组织中MDA、NO无明显差异。(4)结论纳米硫化镉可引起小鼠贫血、体内炎症反应以及肝脏损伤,且纳米材料晶面能越高,损伤作用越强。Objective To study the damage effects on mice hematological indexs and liver tissue induced by two kinds nano-cadmium sulfide materiasls(Nano-CdS)with different crystal surface energy.Methods 24 Kunming mice were selected and divided into three groups with 8 mice in each group,which were respectively the control group,CdS1 group and CdS2 group.The mice in the CdS1 group and CdS2 group were injected intraperitoneally with nano-CdS1 and nano-CdS2 suspensions(0.2mL)through intraperitoneal injection at the doses of 20mg/kg·body weight respectively,and the mice were treated with 0.2mL normal saline in control group,once a week for 4 consecutive weeks.After the exposure,the changes of hematological parameters and aspartate aminotransferase(AST)and alanine aminotransferase(ALT)in serum were measured,and the AST/ALT ratio was calculated in each group.The liver pathological changes were observed by HE staining.The contents of Malondialdehyde(MDA)and Nitric oxide(NO)in serum and liver tissue were detected.Results Compared with control group,the indexs of hemoglobin content,hematocrit and mean corpuscular volume decreased,while the white blood cells,platelets and the ratio of AST/ALT in the CdS2 group increased significantly(P<0.05).The liver tissue in the CdS1 and CdS2 groups had pathological damages in varying degrees,and the injuries in CdS2 group were more serious.Compared with CdS1 group,the contents of MDA and NO in serum in the CdS2 group significantly increased(P<0.05);There were no significant differences between MDA and NO contents in liver tissue of mice.Conclusion Nano-CdS can cause anemia,inflammatory reaction and liver damage in mice.The higher the crystal surface energy of nano-materials,the stronger the damage effect.
