摘  要:目的应用免疫组化(IHC)法比较分析3种不同克隆号间变性淋巴瘤激酶(ALK)抗体在炎性肌纤维母细胞肿瘤(IMT)中的表达。方法选取27例IMT石蜡样本,运用3种不同克隆号的ALK抗体:D5F3(Ventana+Bench Mark)、1A4 (OriGene)及ALK-1(Dako),分别检测在IMT中的表达,并结合FISH检测结果进行比较。结果3种抗体阳性率为D5F3和1A4均为51.9%(14/27),ALK-1为29.6%(8/27)。阳性病例中,D5F3和1A4阳性细胞所占百分比高,染色强度主要为强染色;而ALK-1阳性细胞数量及染色强度均具有显著异质性。11例行FISH检测,D5F3和1A4与FISH结果符合率100%(11/11),ALK-1与FISH结果符合率81.8%(9/11)。抗体D5F3、1A4及ALK-1的敏感性分别为100%(8/8)、100%(8/8)及75%(6/8),三者特异性均为100%(3/3)。结论 3种不同克隆号ALK抗体用于检测IMT时,D5F3与1A4性能相似,均优于ALK-1。Objective To compare and analyze the performance of three different clones of ALK antibody for detecting the expression of ALK protein in Inflammatory myofibroblastic tumor(IMT) by immunohistochemistry(IHC). Methods Three different clones ALK antibodies,ALK D5F3(Ventana),ALK 1A4(OriGene),and ALK-1(Dako) were used to detect the expression of ALK protein in 27 paraffin samples of IMTs,and the results of IHC staining were compared with each other,with the FISH results used as a reference. Results The positive rate was 51. 9%(14/27) for D5F3 and 1A4,and 29. 6%(8/27) for ALK-1. Among the positive cases of D5F3 and 1A4,the percentage of positive cells was high and the staining intensity was strong,but ALK-1 was significantly heterogeneous. FISH was performed in 11 of 27 studied IMTs,the consistency of D5F3 and 1A4 with FISH results was 100%(11/11),and ALK-1 was 81. 8%(9/11). The sensitivity of antibodies D5F3,1A4,and ALK-1 was 100. 0%(8/8),100. 0%(8/8),and 75. 0%(6/8),and the speciticity was all 100%(3/3). Conclusions The performance of D5F3 is similar to 1A4 and both are superior to ALK-1 for detecting ALK protein in IMT by IHC. This study provides reference data for pathologists in the selection of different clones of ALK antibodies for detecting ALK protein in IMT by IHC.
