探究miR-27与IL-10在晚期乳腺癌患者治疗前后的表达及临床意义

摘　　要：目的:探究miR-27与IL-10在晚期乳腺癌患者治疗前后的表达及临床意义。方法:收集2014年5月~2015年9月于本院治疗的晚期乳腺癌患者60例(观察组),另收集同期健康志愿者50例(对照组),分别采用qRT-PCR与ELISA检测并比较2组患者血清中miR-27与IL-10表达;比较观察组患者治疗前后miR-27与IL-10表达的变化;根据临床疗效将患者分为疗效优秀组与疗效较差组比较2组患者治疗前miR-27与IL-10表达,绘制受试者工作曲线(ROC)分析治疗前miR-27与IL-10的表达在疗效预测中的价值;对患者进行3年随访并绘制生存曲线。根据治疗前miR-27与IL-10表达中位值分为高表达组和低表达组,观察miR-27与IL-10高、低表达在患者生存中的价值,分析患者临床疗效。结果:miR-27在对照组受试者血清中表达明显低于观察组,而IL-10表达明显高于观察组(P<0.001);观察组患者通过治疗后血清中miR-27表达明显降低,而IL-10表达明显升高(P<0.001);疗效优秀组患者血清中miR-27表达明显低于疗效较差组,而IL-10表达明显高于疗效较差组(P<0.05);绘制ROC曲线发现miR-27与IL-10曲线下面积分别为0.719与0.685,患者3年存活率为50%,miR-27低表达组患者3年存活率明显高于高表达组患者,而IL-10低表达患者3年生存率明显低于高表达组患者(P<0.05)。结论:miR-27与IL-10可作为晚期乳腺癌患临床疗效与生存的观察指标。Objective:To investigate the expression and clinical significance of miR-27 and IL-10 in advanced breast cancer patients before and after treatment.Methods:60 patients with advanced breast cancer who were treated in our hospital from May 2014 to September 2015(observation group) and 50 volunteers with physical examination at the same time(control group) were collected.Expression of miR-27 and IL-10 in serum was detected and compared by qRT-PCR and ELISA,respectively.Changes of miR-27 and IL-10 expression before and after treatment of patients were observed.According to the clinical efficacy,patients were divided into excellent curative effect group and poor curative effect group to compare expression of miR-27 and IL-10 between the two groups before treatment.Working curve of the subjects was drawn by(ROC) to analyze the predictive value of expression of miR-27 and IL-10 in curative effect before treatment,and the survival curve was drawn by 3-year follow-up.According to the median expression of miR-27 and IL-10 before treatment,patients were divided into high and low expression groups.Survival values of miR-27 and IL-10 expressions in patients were observed,and the clinical efficiency of patients was analyzed.Results:Expression of miR-27 in serum of control group was significantly lower than that in observation group,while expression of IL-10 in patients in control group was significantly higher than that in observation group(P<0.001).Expression of miR-27 in serum of patients in observation group was significantly lower than that in control group after treatment,while expression of IL-10 was significantly increased(P<0.001).Expression of miR-27 in serum of patients with excellent curative effect was significantly lower than patients with poor curative effect group,while expression of IL-10 was significantly higher than that in poor curative effect group(P<0.05),ROC curve showed that area under miR-27 and IL-10 were 0.719 and 0.685,respectively,and the 3-year survival rate was 50%,3-year survival rate of patients
