摘 要:目的探讨小干扰RNA(siRNA)介导染色体空间重组因子(Rsf-1/HBXAP)对子宫内膜癌细胞增殖、凋亡和侵袭的影响。方法选取2016年2月至2018年3月安康市中心医院收治的80例子宫内膜癌患者作为研究对象,采用免疫组织化学法检测80例子宫内膜癌患者癌组织和癌旁正常组织中Rsf-1/HBXAP蛋白表达情况;采用实时荧光定量聚合酶链反应法检测不同子宫内膜癌细胞系(Ishikawa细胞和HEC-1-A细胞)Rsf-1/HBXAP mRNA表达水平;利用脂质体Lipofectamine 2000将siRNA-Rsf-1/HBXAP转染子宫内膜癌细胞作为siRNA-Rsf-1/HBXAP组,同时设置阴性对照组和空白对照组,分别采用细胞计数试剂盒-8法、流式细胞仪及Transwell法检测各组细胞增殖、凋亡和侵袭能力。结果子宫内膜癌癌组织Rsf-1/HBXAP蛋白阳性率高于癌旁正常组织[77.50%(62/80)比18.75%(15/80)](P<0.01)。子宫内膜癌Ishikawa细胞Rsf-1/HBXAP mRNA表达水平高于HEC-1-A细胞(1.34±0.16比1.02±0.28)(P<0.01)。siRNA-Rsf-1/HBXAP组、阴性对照组和空白对照组细胞Rsf-1/HBXAP mRNA分别为0.22±0.04、1.37±0.17和1.30±0.15,3组比较差异有统计学意义(P<0.01)。细胞OD值、细胞凋亡率、侵袭细胞数的组间和时点间存在交互作用(P<0.05),第3天、第5天各组细胞OD值呈升高趋势,siRNA-Rsf-1/HBXAP组各时点间OD值均低于空白对照组和阴性对照组(P<0.05);第3天、第5天空白对照组和阴性对照组细胞凋亡率呈下降趋势,siRNA-Rsf-1/HBXAP组细胞凋亡率呈升高趋势,siRNA-Rsf-1/HBXAP组各时点间细胞凋亡率均高于空白对照组和阴性对照组(P<0.05);第3天、第5天空白对照组和阴性对照组侵袭细胞数呈升高趋势,siRNA-Rsf-1/HBXAP组侵袭细胞数呈下降趋势,siRNA-Rsf-1/HBXAP组各时点间侵袭细胞数均低于空白对照组和阴性对照组(P<0.05)。结论Rsf-1/HBXAP在子宫内膜癌中高表达,沉默Rsf-1/HBXAP可降低癌细胞增殖及侵袭能力,增强其凋亡能力。Objective To investigate the effects of small interfering RNA(siRNA)-mediated chromosome remodeling and spacing factor-1(Rsf-1/HBXAP)on proliferation,apoptosis and invasion of endometrial cancer cells.Methods A total of 80 patients with endometrial cancer admitted to Ankang Central Hospital from Feb.2016 to Mar.2018 were included in the study.The immunohistochemical method was used to detect the expression of Rsf-1/HBXAP in 80 cases of endometrial cancer tissues and normal tissues adjacent to the cancer.Real-time fluorescence quantitative polymerase chain reaction was used to detect the expression of Rsf-1/HBXAP in different endometrial cancer cell lines(Ishikawa cells and HEC-1-A cells).Lipofectamine 2000 was used to transfect siRNA-Rsf-1/HBXAP into endometrial cancer c ells called siRNA-Rsf-1/HBXAP group,and a negative control group and a blank control group were set up.Cell counting kit-8 method,flow cytometry and Transwell method were used to detect the proliferation,apoptosis and invasion of cells in each group.Results The positive rate of Rsf-1/HBXAP protein in the endometrial carcinoma tissues was higher than that in the normal tissues[77.50%(62/80)vs 18.75%(15/80)](P<0.01).The expression of Rsf-1/HBXAP mRNA in Ishikawa cells was higher than that in HEC-1-A cells(1.34±0.16 vs 1.02±0.28)(P<0.01).Rsf-1/HBXAP mRNA in the siRNA-Rsf-1/HBXAP group,negative control group and blank control group was 0.22±0.04,1.37±0.17 and 1.30±0.15,respectively.There was significant difference among the three groups(P<0.01).There were interactions in OD value,apoptosis rate and invasive cell number between groups and time points(P<0.05).On the 3rd and 5th day,the OD value of each group was increased,and the OD value of siRNA-Rsf-1/HBXAP group was lower than that of the blank control group and negative control group at each time point(P<0.05).On the 3rd and 5th day,the apoptosis rate of the blank control group and negative control group decreased,while that of the siRNA-Rsf-1/HBXAP group increased.The apoptosis rate of the si
