摘 要:目的:探讨美托洛尔对大鼠腹腔暴露后肠黏膜屏障的保护作用。方法:建立腹腔暴露大鼠模型,将24只大鼠随机分为三组:对照组、腹腔暴露组和美托洛尔组(腹腔暴露+美托洛尔干预,5 mg/kg),每组8只。观察各组大鼠小肠组织的病理变化,检测体外肠道通透性和血清D-乳酸变化,并通过培养肝、脾、肠系膜淋巴结细菌观察肠道细菌易位情况。结果:腹腔暴露组较对照组大鼠肠黏膜损伤Chiu’s评分,体外肠道通透性,血清D-乳酸和肠道细菌易位明显增加(P<0.05);美托洛尔组较腹腔暴露组大鼠肠黏膜损伤Chiu’s评分,体外肠道通透性,血清D-乳酸和肠道细菌易位明显降低(P<0.05)。结论:美托洛尔能降低腹腔暴露后的肠黏膜损伤,有助于保护腹部手术后的肠黏膜屏障功能。Objective:To investigate the protective effect of metoprolol on intestinal mucosal barrier after open abdomen in rats.Methods:A rat model of open abdomen was established.Twenty-four rats were randomized into three groups:control group,open abdomen group,and metoprolol group(open abdomen plus 5 mg/kg metoprolol intervention),respectively(n=8 each).The terminal ileum was sampled to observe histological changes and to analyze Chiu’s score for damage of intestinal mucosa.Blood was obtained to determinate serum D-lactate;terminal ileum was sampled to determinate in vitro intestinal permeability;liver,spleen and mesenteric lymph nodes were excised for evaluation of bacterial translocation.Results:Compared to control group,open abdomen group showed a significant increase in Chiu’s score for damage of intestinal mucosa,serum D-lactate,in vitro intestinal permeability,and bacterial translocation rate(P<0.05).Compared to open abdomen group,metoprolol group showed a significant decrease in Chiu’s score for damage of intestinal mucosa,serum D-lactate,in vitro intestinal permeability,and bacterial translocation rate(P<0.05).Conclusion:The data suggest that metoprolol could decrease intestinal mucosal damage induced by open abdomen,which is conducive to maintain intestinal mucosal barrier function after abdominal surgery.
