摘 要:文章采用圆红冬孢酵母进行油脂发酵,并研究以其湿菌泥为原料,采用NaOH热辅助破壁进行油脂提取的方法。研究结果表明:当NaOH添加量为0.2 g/g时,样品经过78℃水浴处理3 h并用乙酸乙酯萃取,油脂提取率可达到99.3%;碱热法所得油脂的脂肪酸组成和中性脂含量与酸热法无显著差异。因此,碱热法是一种以湿菌泥为原料直接进行油脂提取的新方法,具有良好的应用前景。In this paper,Rhodosporidium toruloides was used to production lipids,and using NaOH heat-assisted broken cell wall to extract lipids with wet yeast as raw material.The results demonstrate that when the NaOH addition amount was 0.2 g/g,the sample was treated in a 78℃water bath for 3 h and extracted with ethyl acetate,the lipid yield could reach 99.3%;There was no significant difference in the composition of neutral esters and fatty acids between alkali-heat-assisted and traditional acid-heat methods.Therefore,this new extraction method can directly extract lipids from wet yeast,which is good prospects for industrial application production of microbial lipids.
