摘 要:全氟辛基碘烷(PFOI)作为化工中间体,其衍生物的广泛应用使得其具有较高的商业价值。但其环境暴露的毒性效应研究甚少。以乳腺癌细胞(MCF-7)为离体模型研究PFOI暴露对细胞迁移能力的影响以及对长链非编码RNA MALAT-1的调节;以斑马鱼胚胎为活体模型来研究斑马鱼胚胎在受到PFOI急性暴露后的胚胎毒性效应,以及对MALAT-1的调节作用。结果表明,PFOI暴露后MCF-7细胞的迁移能力提高,且细胞中MALAT-1基因被诱导高表达;在活体实验中,PFOI(50μmol·L^-1和100μmol·L^-1)暴露导致斑马鱼胚胎发育迟缓,胚胎自发运动因不受神经调节控制而更加活跃,500μmol·L^-1 PFOI暴露组中孵化率在显著性下降,且在幼鱼脑部集中表达的MALAT-1基因被诱导上调。以上研究结果表明,MALAT-1参与了PFOI诱导癌细胞迁移和自发运动过度活跃等毒性效应的调节作用。The derivatives of 1-iodoperfluorooctane(PFOI)are widely used,so PFOI as an intermediate in chemical industry,has a higher commercial value.However,the toxic effects of PFOI was not well studied.We used breast cancer cell line(MCF-7)as an in vitro model to study the effects of PFOI exposure on cell migration and the regulation of long non-coding RNA MALAT-1.Zebrafish embryos were used as an in vivo model to study the em-bryo toxicity and the change of MALAT-1 upon PFOI acute exposure.The results showed that the migration ability of MCF-7 cells was improved after PFOI exposure,and the high expression of MALAT-1 gene was induced in MCF-7 cells.In vivo,PFOI(50μmol·L^-1 and 100μmol·L^-1)exposure resulted in delayed embryonic develop-ment,and the spontaneous movement of zebrafish embryos became more active because they were not controlled by neuroregulation.The hatching rate of zebrafish embryos was significantly decreased in the group exposed to PFOI(50μmol·L^-1 and 100μmol·L^-1).It also induced the upregulation of MALAT-1 gene in the brain of zebrafish larvae.These results suggest that MALAT-1 is involved in the regulation of toxic effects such as cell migration and spontaneous hyperactivity induced by PFOI.
