摘 要:目的研究鼻咽癌组织中缝隙连接蛋白43(Cx43)、Cx26的表达与患者临床病理特征及肿瘤侵袭活力的相关性。方法收集2014年3月—2017年12月在我院经病理活检确诊的鼻咽癌组织60例及同期经病理活检证实为正常的鼻咽组织42例,检测Cx43、Cx26及侵袭基因的表达水平。结果鼻咽癌组织中Cx43、Cx26的阳性表达率分别为35.00%、43.00%,均明显低于正常鼻咽组织(71.43%、76.19%)(P<0.05);且T3-T4期、临床分期III-IV期、有淋巴结转移的鼻咽癌组织中Cx43、Cx26的阳性表达率明显低于T1-T2期、临床分期I-II期、无淋巴结转移的鼻咽癌组织(P<0.05)。鼻咽癌组织中MMP-2、MMP-3、MMP-9、Cat-D的mRNA表达量明显高于正常鼻咽组织(P<0.05);且Cx43、Cx26阳性鼻咽癌组织中MMP-2、MMP-3、MMP-9、Cat-D的mRNA表达量明显低于Cx43、Cx26阴性鼻咽癌组织(P<0.05)。结论Cx43、Cx26低表达可能与鼻咽癌患者的临床病理特征改变、肿瘤侵袭活力增强有关,并参与了鼻咽癌的发生发展过程。Objective To analyze the correlation of the expressions of connexin 43(Cx43)and Cx26 in nasopharyngeal carcinoma with the clinicopathological characteristics and tumor invasive activity.Methods Nasopharyngeal carcinoma tissues and normal nasopharyngeal tissues confirmed by pathological biopsy between March 2014 and December 2017 were collected.The expression levels of Cx43,Cx26 and invasive genes were determined.Results The positive expression rates of Cx43 and Cx26 were significantly lower in the nasopharyngeal carcinoma than in the normal nasopharyngeal tissues(35.00%vs.71.43%,43.00%vs.76.19%,respectively)(P<0.05).The positive expression rates of Cx43 and Cx26 in nasopharyngeal carcinoma tissues of T3-T4 stage,clinical stage III-IV,or lymph node metastasis were significantly lower than those in T1-T2 stage,clinical stage I-II or non-lymph node metastasis nasopharyngeal carcinoma(P<0.05).The mRNA expression levels of MMP-2,MMP-3,MMP-9 and Cat-D in nasopharyngeal carcinoma tissues were significantly higher than those in normal nasopharyngeal tissues(P<0.05).Moreover,the mRNA expression levels of MMP-2,MMP-3,MMP-9 and Cat-D in nasopharyngeal carcinoma tissues with positive expression of Cx43 and Cx26 were significantly lower than those in nasopharyngeal carcinoma tissues with negative expression of Cx43 and Cx26(P<0.05).Conclusion The low expression of Cx43 and Cx26 in nasopharyngeal carcinoma may be correlated with the changes of clinicopathological features and the enhancement of invasive activity of the tumor,and involved in the occurrence and development of nasopharyngeal carcinoma.
