摘　　要：目的:探讨Ca^2+信号通路对大鼠舌黏膜鳞癌形成的影响。方法:将70只SD大鼠随机分为空白对照组(A)和实验组。随机数字法抽取8只SD大鼠纳入对照组,不做任何处理。其余为总实验组,分别在第4周、8周、12周、16周、20周、24周、28周,在总实验组中随机抽取8只SD大鼠分别记录为B组(4周)、C组(8周)、D组(12周)、E组(16周)、F组(20周)、G组(24周)、H组(28周)。抽取的SD大鼠处死后,切取舌组织并提取总RNA,Genechip Rat Genome 230 2.0芯片测定舌组织mRNA表达,分析Ca^2+信号通路基因表达变化情况。结果:Ca^2+信号通路中GPCR、ROC、CD38、ADCY、SERCA、RYR、PKC等关键基因均上调;下调的有NCX、PLCβ、PLN、IP3R、CAMK等基因。结论:Ca^2+信号通路可能是调控舌黏膜鳞癌形成的重要信号通路之一。Objective:To investigate the effect of Ca2+signaling pathway on the formation of tongue mucosal squamous cell carcinoma in rats.Methods:70 SD rats were randomly divided into blank control group(A)and experimental group.Eight SD rats were selected by random number method and included in the control group without any treatment.The rest were experimental groups at week 4,week 8,week 12,week 16,week 20,week 24,and week 28,respectively.Eight SDS randomly selected from the experimental group were recorded as group B(4 weeks),group C(8 weeks),group D(12 weeks),group E(16 weeks),group F(20 weeks),group G(24 weeks),and group H(28 weeks).After the SD rats were sacrificed,the tongue tissue was cut and the total RNA was extracted.Genechip Rat Genome 2302.0 chip was used to determine the mRNA expression in the tongue tissue and analyze the gene expression changes of Ca2+signaling pathway.Results:GPCR,ROC,CD38,ADCY,SERCA,RYR,PKC and other key genes were up-regulated in Ca2+signaling pathway.Down-regulated NCX,PLC,PLN,IP3R,CAMK and other genes.Conclusion:Ca2+signaling pathway may be one of the important signaling pathways that regulate the formation of squamous cell carcinoma of the lingual mucosa.
