摘 要:目的对比分析山葡萄酒与欧亚葡萄酒对大鼠肾上腺嗜铬瘤细胞(pheochromocytoma cells,PC-12)氧化性损伤的保护作用强弱,并探讨其可能的原因。方法建立H2O2诱导PC-12细胞氧化损伤模型,采用噻唑蓝(thiazolyl blue tetrazolium bromide,MTT)法测定细胞存活率,确定H2O2的最佳作用浓度;采用不同浓度的葡萄酒对PC-12细胞预处理24 h,确定对PC-12细胞无影响的浓度后,MTT法比较不同品种和不同年份的山葡萄酒和欧亚葡萄酒对H2O2致PC-12细胞存活率下降的改善作用的差别;并检测细胞内谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、超氧化物歧化酶(superoxide dismutase,SOD)活性及丙二醛(malondialdehyde,MDA)的含量;测定不同品种、不同年份的山葡萄酒和欧亚葡萄酒中总多酚、总黄酮、总原花青素和总花色苷的含量,探究葡萄酒对PC-12细胞氧化性损伤的保护作用与多酚成分之间的关系。结果与模型组相比,山葡萄酒和欧亚葡萄酒均能改善H2O2致PC-12细胞存活率的降低。山葡萄酒较欧亚葡萄酒能更显著地提高PC-12细胞存活率,且其提高氧化损伤的PC-12细胞内GSH-Px、SOD活性,降低MDA含量的作用均显著优于欧亚葡萄酒,在高浓度时保护作用差距更显著。山葡萄酒中多种多酚类成分含量均显著高于欧亚葡萄酒。相关性分析表明,葡萄酒对PC-12细胞氧化性损伤的保护作用与其总多酚、总黄酮、总原花青素和总花色苷含量呈正相关。结论山葡萄酒比欧亚葡萄酒对H2O2所致PC-12细胞损伤具有更显著的保护作用。山葡萄酒多酚类成分含量高,可更好地增强抗氧化酶活力和减轻脂质过氧化反应,这可能是山葡萄酒对细胞氧化应激保护作用高于欧亚葡萄酒的原因。Objective To compare the effects of wines of Vitis amurensis and Vitis vinifera against oxidative stress in pheochromocytoma cells(PC-12).Methods H2O2-induced oxidative stress model in PC-12 cells was established.Survival rates were detected by MTT assay to determine the optimal concentration of H2O2.Cells were treated with various concentrations of wine for 24 h.Compared the difference of survival rates in H2O2-induced PC-12 cells that were processed by wines of Vitis amurensis or Vitis vinifera with different varieties and different years by MTT.The cell viability results were calculated as the percentages of reduced MTT.Wine of 2016 was chosen to further evaluate the superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)activities and the malonic dialdehyde(MDA)content in PC-12 cells.The contents of total phenolics,total flavonoids,total proanthocyanins and total anthocyanins in wines of Vitis amurensis and Vitis vinifera with different concentrations and varieties were measured by traditional chemical analysis methods.Results Both wines of Vitis amurensis and Vitis vinifera had protective effects against oxidative stress in PC-12 cells,whereas the effect of Vitis amurensis wine was significantly stronger than that of Vitis vinifera wine.The effects of pretreatment with Vitis amurensis wine on both the ecnhancement of the activities of SOD and GSH-Px,and the reduction of MDA levels,were significantly stronger than pretreatment with Vitis vinifera wine in a high concentration.The contents of determined kinds of polyphenols in Vitis amurensis wine were significantly higher than that in Vitis vinifera wine.There was a close relationship between total phenolics,total flavonoids,total proanthocyanins,total anthocyanins and the antioxidant activity of wine according to correlation analysis.Conclusion The protective effect of Vitis amurensis wine against oxidative stress in PC-12 cells is significantly stronger than that of Vitis vinifera wine.The high content of polyphenols can enhance the activity of antioxida
