摘  要:目的观察加味附子理中汤预防性给药对缺血再灌注急性肾损伤大鼠肾功能、氧化应激及细胞凋亡的影响。方法采用数字随机表法将45只SD雄性大鼠随机分为假手术组、缺血再灌注组、缺血再灌注+药物组,每组15只。缺血再灌注+药物组于术前连续7 d灌胃加味附子理中汤[10 mL/(kg·d)],假手术组、缺血再灌注组给予等容量的生理盐水。建立缺血再灌注模型24 h后,采用全自动生化分析仪检测血清肌酐(SCr)、尿素氮(BUN)水平,比色法测定肾组织中超氧化物歧化酶(SOD)、丙二醛(MDA)表达水平,Western blot法检测肾组织中Caspase-3、PARP、Bax表达水平。结果缺血再灌注组BUN、SCr水平明显高于假手术组(P均<0.05),缺血再灌注+药物组BUN、SCr水平均明显低于缺血再灌注组(P均<0.05)。缺血再灌注组肾组织中MDA、Caspase-3、PARP、Bax表达水平均明显高于假手术组(P均<0.05),SOD表达水平明显低于假手术组(P<0.05);缺血再灌注+药物组肾组织中MDA、Caspase-3、PARP、Bax表达水平均明显低于缺血再灌注组(P均<0.05),SOD表达水平明显高于缺血再灌注组(P均<0.05)。结论预防性使用加味附子理中汤可以改善缺血再灌注急性肾损伤大鼠的肾功能,作用可能与抑制氧化应激及调控细胞凋亡通路有关。Objective It is to observe the effects of modified Fuzi Lizhong Decoction on renal function,oxidative stress and apoptosis in rats with acute kidney injury after ischemia reperfusion(IR).Methods A total of 45 SD male rats were randomly divided into 3 groups:sham group,IR group and IR+medicine group according to random number method,15 rats in each group.The IR+medicine group was continuously treated with modified Fuzi Lizhong Decoction[10 mL/(kg·d)]for 7 days by gavage before the operation,the sham operation group and the IR group were treated with equal volume of normal saline.24 hours after the IR models were established,the levels of serum creatinine(SCr)and urea nitrogen(BUN)were detected by an automatic biochemical analyzer,and the expression levels of superoxide dismutase(SOD)and malondialdehyde(MDA)in kidney tissue were determined by colorimetry,the expression levels of Caspase-3,PARP and Bax in kidney tissue was detected by Western blot method.Results The levels of BUN and SCr in the IR group were significantly higher than those in the sham operation group(all P<0.05),and the levels of BUN and SCr in the IR+medicine group were significantly lower than those in the IR group(all P<0.05).The expression levels of MDA,Caspase-3,PARP,and Bax in the renal tissue of the IR group were significantly higher than those in the sham operation group(all P<0.05),and the SOD expression level was significantly lower than that in the sham operation group(P<0.05);the expression levels of MDA,Caspase-3,PARP and Bax in the renal tissues of the IR+medicine group were significantly lower than those in the IR group(all P<0.05),and the expression level of SOD was significantly higher than that in the IR group(all P<0.05).Conclusion Prophylactic administration of Fuzi Lizhong Decoction can improve renal function in rats with acute renal injury due to ischemia-reperfusion,which may be related to inhibiting oxidative stress and regulating apoptotic pathways.
