摘 要:目的研究Claudin-1与cyclinB1在下咽鳞状细胞癌及癌旁组织中的表达情况,分析它们与下咽鳞状细胞癌各临床因素及患者预后之间的关系。方法 (1)选取97例下咽鳞状细胞癌组织学标本及90例癌旁正常黏膜组织标本,采用免疫组化的方法比较Claudin-1和cyclinB1在下咽鳞状细胞癌及正常黏膜组织中表达的差异,分析两者在下咽鳞状细胞癌中的表达与临床因素及患者预后之间的关系;(2)选取30位下咽鳞状细胞癌患者术后的癌组织及癌旁正常黏膜,采用RT-PCR方法,检测Claudin-1和cyclinB1的表达并定量分析,探讨两者的相关性。结果 (1)Claudin-1与cyclinB1在下咽鳞状细胞癌中的阳性表达率均明显高于癌旁组织(P<0.05);(2)Claudin-1蛋白表达与肿瘤的分化程度(P=0.004)和淋巴结转移(P=0.026)有关,与患者的生存率相关(P=0.003);(3)CyclinB1蛋白表达与肿瘤的分化程度(P<0.001)有关;(4)Claudin-1与cyclinB1呈明显正相关关系(r=0.482,P<0.001)。结论 Claudin-l和cyclinB1具有协同作用,二者的高表达可能促进了下咽癌的发生与发展。Objective To investigate the expressions of claudin-1 and cyclinB1 in hypopharyngeal squamous cell carcinoma(HSCC) and its adjacent tissues, and to analyze their relationship with clinical factors and prognosis of patients with HSCC.Methods(1)The histological specimens of 97 cases with complete clinical pathological data of the patients with HSCC were selected as the research object, and 90 cases of adjacent normal mucosa tissues as the control group.The expressions of claudin-1 and cyclinB1 in HSCC and normal mucosa were analyzed by immunohistochemistry.It was analyzed for the relationship between the clinical factors and the prognosis of patients.(2)The expressions of claudin-1 and cyclinB1 in 30 cases of HSCC and its adjacent tissues were analyzed by RT-PCR, and to explore the correlation between them.Results(1)Both Claudin-1 and cyclinB1 were highly expressed in HSCC compared to adjacent tissues(P<0.05).(2)Claudin-1 protein expression was correlated with tumor differentiation(P=0.004)and lymph node metastasis(P=0.026). Kaplan-Meier analysis showed that Claudin-1 was associated with patient survival(P=0.003).(3)The expression of cyclinB1 was correlated with the differentiation degree of tumor(P<0.001). Claudin-1 was positively correlated with cyclinB1(P<0.001).(4)The relationship between claudin-1 and cyclinB1 was significantly positive correlation(r=0.482,P<0.001).Conclusion The highly expressions of claudin-l and cyclinB1 with synergistic effects may contribute to the carcinogenesis and development of hypopharyngeal carcinoma.
