摘　　要：目的初步探讨抑癌基因RBM 5和凋亡抑制蛋白survivin在前列腺癌(PCa)组织中表达的意义。方法采用免疫组织化学S-P,半定量逆转录PCR(RT-PCR)和Western blot方法分别检测正常前列腺(NP)组织、良性前列腺增生(BPH)组织和PCa组织中RBM5和survivin蛋白的表达情况,并分析其与临床病理特征之间的相关性。结果RBM5在NP、BPH和PCa组织中的阳性表达率分别是83.33%、77.13%和11.42%,组间比较,差异有显著统计学意义(P<0.01)。survivin的阳性表达率分别是6.67%、15.71%和84.38%,组间对比,差异明显(P<0.01)。RTPCR结果显示,RBM 5蛋白在NP、BPH组织中高表达,在PCa组织中表达缺失,组间比较差异明显(P<0.01);survivin在NP、BPH中几乎无表达,在PCa中则高表达,组间比较差异有显著统计学意义(P<0.01)。Western blot对RBM5和survivin蛋白在NP、BPH和PCa组织中表达定量分析,进一步证实PCa组织与NP和BPH组织表达差异显著(P<0.01)。RBM5和survivin的表达水平与PCa细胞的分化程度和临床分型存在相关性(P<0.05)。结论RBM 5和survivin的表达程度极有可能与PCa的发生发展密切相关。增强RBM5表达的同时沉默survivin的双靶点用药有望提高分子靶向治疗去势抵抗的雄激素非依赖性PCa患者的预后。Objective To investigate the expression and significance of tumor suppressor gene RBM5and survivin in prostate cancer(PCa)tissues.Methods Immunohistochemical,semi-quantitative reverse transcriptional PCR(RTPCR)and Western blot were used to detect the expressions of RBM5and survivin proteins in normal prostate(NP)tissues,benign prostatic hyperplasia(BPH)tissues and PCa tissues,respectively,and to analyze their correlation with clinicopathological characteristics.Results The positive expression rates of RBM5in NP,BPH tissues and PCa tissues were 83.33%,77.13%and 11.42%,respectively.The difference between groups was statistically significant(P<0.01).The positive expression rates of survivin were 6.67%,15.71%and 84.38%,respectively.Survivin,almost no expression in the NP and BPH is highly expressed in the PCa,compare the differences between groups have significant statistical significance(P<0.01),Western blot of RBM5and survivin protein in the NP,BPH and PCa tissues can express quantitative analysis,further confirmed the PCa with NP and BPH tissue expressed significant difference(P<0.01).RBM5and the expression of survivin levels and PCa cell differentiation degree correlation(P<0.05).Conclusion The degree of the expression of survivin and RBM5are closely related to the development of PCa likely increase RBM5expression level at the same time the double targets for drugs inhibit the expression of survivin are helpful to improve the level of molecular targeted therapy.
