摘 要:目的观察肾细胞癌中Holliday交叉识别蛋白基因和蛋白表达的改变。方法肾癌A498细胞、os-rc-2细胞作为实验组,人肾小管上皮细胞HK2作为对照组,实时荧光定量聚合酶链反应分析Holliday交叉识别蛋白在实验组和对照组的基因表达情况,蛋白印迹分析其蛋白表达情况。结果与对照组人肾近曲小管上皮细胞HK2细胞相比较,os-rc-2和A498肾癌细胞中Holliday交叉识别蛋白基因和蛋白表达水平均明显下降(P<0.01),在A498细胞中降低的更为明显。结论Holliday交叉识别蛋白基因和蛋白表达水平在肾细胞癌中均明显降低,可能作为一种分子靶标在肾细胞癌的诊断,治疗和预后判断中发挥重要的作用。Objective To observe the changes of Holliday Junction Recognition Protein gene and protein expression in renal cell carcinoma.Methods Renal cell cancer A498cells and os-rc-2cells were used as experimental group.Human renal tubular epithelial cells HK2were used as control group.Real-time quantitative polymerase chain reaction was used to analyze the gene expression of Holliday Junction Recognition Protein in experimental group and control group.Western blot analysis of its protein expression.Results Compared with the control group of renal proximal tubular epithelial cells HK2cells,the expression levels of Holliday Junction Recognition Protein gene and protein in os-rc-2and A498renal cancer cells were significantly decreased(P<0.01),and decreased in A498cells.obvious.Conclusion The protein and gene expression level of Holliday Junction Recognition Protein are significantly decreased in renal cell carcinoma,which may play an important role in the diagnosis,treatment and prognosis of renal cell carcinoma.
