摘 要:目的检测T细胞免疫球蛋白黏连蛋白-3(TIM-3)在宫颈癌患者外周血T细胞中的表达,探讨TIM-3与宫颈癌发生发展的相关性。方法应用流式细胞仪检测CD4^+T细胞,CD8+细胞及TIM-3在外周血不同类型T细胞表面的表达。收集21例对照组、15例宫颈上皮内瘤变(CIN)患者和24例子宫颈癌患者样本,进一步分析TIM-3与宫颈癌临床病理参数的关系。结果宫颈癌及CIN患者较非宫颈病变患者外周血的CD4^+T及CD8^+T细胞明显减少,并且CD4^+T及CD8^+T细胞表面的TIM-3表达量明显增高,有统计学意义,CIN与宫颈癌相比较差异无统计学意义。与Ⅰ~Ⅱ期比较,TIM-3在宫颈癌Ⅲ~Ⅳ期外周血CD4^+T及CD8^+T细胞中的表达显著增加,差异有统计学意义(P<0.05)。不同临床因素分析结果显示,TIM-3在外周血T细胞中的表达与年龄、淋巴结阳性、肿瘤大小及鳞状细胞癌抗原高低无相关性(P>0.05)。结论TIM-3与宫颈癌的发生发展有关,可能成为宫颈癌的一种新的免疫治疗靶点。Objective In this study,we aimed to investigate the expression of T cell immunoglobulin and mucin domain-containing protein 3(TIM-3)in T cells from the peripheral blood of cervical cancer patients,and to explore the relationship between TIM-3 and the development of cervical cancer.Methods CD4^+T cells and CD8^+T cells in the peripheral blood of patients with cervical cancer and CIN were significantly reduced compared with those from patients without cervical lesions,and the expression levels of TIM-3 on the surface of CD4^+T and CD8^+T cells were significantly increased,in a statistically significant fashion;on the other hand,no statistically significant differences were observed in samples from CIN versus cervical cancer patients.Compared with stageⅠtoⅡ,TIM-3 expression in the levels of peripheral blood of CD4^+T and CD8^+T cells were significantly increased in stageⅢtoⅣcervical cancer patients(P<0.05).Analysis of different clinical factors showed that the expression of TIM-3 in peripheral blood T cells was not correlated with age,lymph node positivity,tumor size,or squamous cell carcinoma antigen level(P>0.05).Conclusion TIM-3 may affect the development and progression of cervical cancer;our data may support the use of TIM-3 for the development of novel immunotherapeutic approaches effective against cervical cancer.
