摘  要:目的制备高活性EB病毒衣壳抗原P18-23融合抗原并初步评价该抗原在鼻咽癌诊断中的应用价值。方法利用分子生物学技术进行克隆表达获得纯化抗原,采用酶联免疫吸附试验初步抗原的检测性能。结果获得了高效原核表达的衣壳抗原P18-23融合抗原,建立了基于该抗原的IgA间接ELISA方法,检测灵敏度和特异度分别为89.77%和86.32%,抗VCA-IgA诊断NPC的AUC值为0.883。结论原核表达的衣壳抗原P18-23融合抗原具有良好的检测性能,利用该抗原建立的VCA-IgA间接ELISA检测方法可以很好地区分鼻咽癌患者和健康对照,适合用于NPC的鉴别诊断。Objective The Epstein-Barr virus capsid antigen P18-23 fusion antigen were prepared and its application value in the diagnosis of nasopharyngeal carcinoma was preliminarily evaluated.Methods The antigens were cloned and expressed using molecular biology technology,and their detection performance was preliminarily evaluated by enzyme-linked immunosor-bent assay(ELISA).Results The EB virus capsid antigen p18-23 fusion antigen was highly prokaryotic expressed in E.coil,and a VCA-IgA indirect ELISA method based on this antigen was established.The detection sensitivity and specificity were 89.77%and 86.32%respectively,and the AUC value of VCA-IgA was 0.883.Conclusion The recombinant virus capsid antigen P18-23 fusion antigen has good detection performance.The indirect ELISA detection method of VCA-IgA established using this antigen can be used to distinguish NPC patients from healthy controls,which is suitable for the differential diagnosis of NPC.
