摘 要:【目的】研究薇菜多糖的单糖组成及其对ABTS+·的清除动力学,为薇菜资源的开发利用提供理论基础。【方法】采用水提醇沉法提取薇菜多糖(POJT),Sevage法脱蛋白后利用SephadexG-200进行分离纯化,得到薇菜多糖主要组分POJT-1,初步分析其单糖组成和结构,同时测定0.2,0.4,0.6,0.8和1.0 mg/mL POJT-1对ABTS+·的清除动力学。【结果】POJT-1的红外光谱显示其具有多糖的特征吸收峰,PMP衍生结合HPLC分离可初步判断POJT-1中可能含有甘露糖、鼠李糖、半乳糖醛酸、葡萄糖、木糖、岩藻糖等单糖组分。不同质量浓度POJT-1清除ABTS+·反应达到平衡的时间存在一定差异,准二级反应动力学能更好地拟合POJT-1清除ABTS+·的反应动力学过程。POJT-1对ABTS+·的清除能力具有剂量效应关系,随着多糖质量浓度的增加,ABTS+·清除速率加快,POJT-1清除ABTS+·的IC 50为0.2737 mg/mL。【结论】薇菜多糖对ABTS+·具有较好的清除效果,且准二级反应动力学能很好地拟合薇菜多糖对ABTS+·的清除作用。【Objective】To improve the development and utilization of Osmanthus japonica Thunb resources,the monosaccharide composition of polysaccharides from O.japonica Thunb was analyzed and its scavenging kinetics on ABTS+·free radicals were investigated.【Method】The polysaccharides of O.japonica Thunb(POJT)were obtained through water extraction and alcohol precipitation.SephadexG-200 was used for separation and purification after deproteinization by Sevage method,and a major component POJT-1 was obtained.The monosaccharides composition and structure of POJT-1 were analyzed and the scavenging kinetics on ABTS+·free radical at concentrations of 0.2,0.4,0.6,0.8 and 1.0 mg/mL POJT-1 were established.【Result】The infrared spectrum of POJT-1 showed characteristic absorption peak,and the PMP derivative combined with HPLC separation could predict preliminarily that POJT-1 may contain mannose,rhamnose,galacturonic acid,glucose,xylose,fucose and other monosaccharide components.The reaction equilibrium time of POJT-1 scavenging ABTS+·free radicals at different mass concentrations had certain differences.The reaction kinetics of POJT-1 scavenging ABTS+·free radicals can be fitted by quasi-secondary reaction kinetics.The scavenging effect of POJT-1 on ABTS+·free radical had a dose-effect relationship.With the increase of polysaccharide concentration,the scavenging rate on ABTS+·free radical increased.The IC 50 of POJT-1 on scavenging ABTS+·free radical was 0.2737 mg/mL.【Conclusion】The polysaccharides of O.japonica Thunb had good scavenging effect on ABTS+·free radical,and the quasi-second-order reaction kinetics could well fit the scavenging effect on ABTS+·free radical.
