摘  要:目的:观察小鼠正常口腔黏膜、口腔黏膜下纤维化(oral submucous fibrosis,OSF)早期、中期、晚期微血管密度(microvessel density,MVD)与胶原堆积的变化关系,探讨MVD变化对小鼠OSF发生发展的作用。方法:采用免疫组化检测小鼠正常口腔黏膜、OSF早期、中期、晚期MVD和Ⅰ型胶原蛋白(collagen typeⅠ,ColⅠ)、Ⅲ型胶原蛋白(collagen typeⅢ,ColⅢ)的变化情况。结果:病变区域MVD出现短暂增加后逐渐下降的趋势,早期组MVD高于正常口腔黏膜和OSF中期、晚期组(P<0.05),中期、晚期组MVD均低于正常口腔黏膜和OSF早期组(P<0.05)。ColⅠ的表达量与MVD变化相反,在正常组、OSF早期、中期、晚期组逐渐增加,差异均有统计学意义(P<0.05),ColⅢ的表达量与MVD变化无直接关系,各组差异均无统计学意义。结论:OSF局部缺血可能会加重ColⅠ堆积,促进纤维化的形成和发展,提示增加局部血运微循环,也许能达到治疗OSF的目的。Objective:To observe the changes of microvessel density(MVD)and collagen accumulation in normal oral mucosa and oral submucous fibrosis(OSF)in the early,middle and late stagesmice of mice,and to explore the effect of MVD on the development of mouse OSF.Methods:Immunohistochemistry was used to detect the changes of MVD and collagen typeⅠ(ColⅠ)and collagen typeⅢ(ColⅢ)in normal oral mucosa,OSF in the early,middle and late stage.Results:The MVD in the lesion area decreased gradually after a brief increase.The MVD in the early stage group was higher than that in the normal group and the middle and late stage group(P<0.05),but the middle and late groups were lower than that in the normal group and the early group(P<0.05).The expression level of ColⅠwas opposite to that of MVD,which increased gradually in each group,and the difference was statistically significant(P<0.05).In the normal group and the early,middle and late stages of OSF,the difference was statistically significant(P<0.05).The expression level of ColⅢwas not directly related to the change of MVD,and the difference of each group was not statistically significant.Conclusion:Ischemia can aggravate the accumulation of ColⅠand promote the formation and development of fibrosis,suggesting an increase in local blood microcirculation,which may achieve the purpose of treating OSF.
