摘  要:目的:探究姜黄素对口腔黏膜下纤维化(oral submucosal fibrosis,OSF)模型大鼠口腔黏膜下纤维化的改善作用。方法:将建模成功的42只OSF模型大鼠随机分为模型组、雷公藤多甙组、姜黄素组各14只。姜黄素组大鼠采用400 mg/kg的姜黄素灌胃,雷公藤多甙组采用30 mg/kg雷公藤多甙液灌胃,模型组、正常组不做任何干预。在4周后观察大鼠颊黏膜颜色、质地变化、测量张口度、颊黏膜评分及颊黏膜组织中Ⅰ型胶原、Ⅲ型胶原及ⅠFN-γmRNA、Wnt/β-catenin信号通路因子表达量。结果:雷公藤多甙组、姜黄素组张口度高于模型组、低于正常组,颊黏膜评分、颊黏膜组织中Ⅰ型胶原、Ⅲ型胶原、ⅠFN-γ、Wnt1、β-catenin、TGF-β1 mRNA表达量均低于模型组、高于正常组(P<0.05)。姜黄素组张口度高于雷公藤多甙组,黏膜评分、颊黏膜组织中Ⅰ型胶原、Ⅲ型胶原、ⅠFN-γ、Wnt1、β-catenin、TGF-β1 mRNA表达量均低于雷公藤多甙组(P<0.05)。结论:姜黄素经作用于Wnt/β-catenin信号通路,调控通路因子及Ⅰ型胶原、Ⅲ型胶原及ⅠFN-γ表达,改善大鼠口腔黏膜下纤维化,进而阻止纤维化进一步发展。Objective:To investigate the effect of curcumin on the improvement of oral submucosal fibrosis(OSF)in rats.Methods:Totally 42 OSF model rats were randomly divided into model group,Tripterygium wilfordii glycoside group and curcumin group.The rats in curcumin group were perfused with 400 mg/kg curcumin,and the rats in Tripterygium glycoside group were perfused with 30 mg/kg Tripterygium glycoside solution.The rats in model group and normal group were not intervened.After 4 weeks,the changes of buccal mucosa color and texture,the measurement of mouth opening,buccal mucosa score and the expression of type I collagen,type III collagen,IFN-γmRNA and Wnt/β-Catenin signal pathway factor were observed.Results:The open mouth of Tripterygium wilfordii polyglycoside group and curcumin group was higher than that of model group and lower than that of normal group.The expression of type I collagen,type III collagen and IFN-γ,Wnt1,β-catenin,TGF-β1 mRNA in buccal mucosa was lower than that in model group and higher than that in normal group(P<0.05).The expression of type I collagen,type III collagen and IFN-γ,Wnt1,β-catenin,TGF-β1 mRNA in curcumin group than that in Tripterygium wilfordii group(P<0.05).Conclusion:Curcumin acting on Wnt/β-catenin signaling pathway,regulates the expression of pathway factors and collagen I,collagen III and IFN-γ,improves the suboral fibrosis in rats,and then prevents the further development of fibrosis.
