摘 要:目的铁死亡是不同于细胞自噬、凋亡和坏死的一种新的细胞死亡方式,与各种人类疾病密切相关。肿瘤的发病率逐年增加,但治疗效果并不乐观,因此应积极探索新的抗肿瘤治疗靶点。本研究总结了铁死亡在肿瘤中的作用机制以及化疗、放疗、分子靶向治疗、免疫治疗、中药抗肿瘤药物和纳米抗肿瘤药物中以铁死亡作为潜在治疗靶点的研究现状和进展。方法以"铁死亡、化疗、放疗、分子靶向治疗和免疫治疗"等作为关键词,应用PubMed及CNKI期刊全文数据库检索系统,检索2012-01-2020-01的相关文献。共检索到英文文献120篇,中文文献5篇。纳入标准:(1)铁死亡发现史;(2)铁死亡在肿瘤中的发生机制;(3)以铁死亡作为潜在靶点的抗肿瘤治疗方法。排除标准:(1)综述类文献;(2)研究资料缺失。根据纳入和排除标准,共纳入分析文献50篇。结果铁死亡作用机制与细胞内铁代谢水平、脂质过氧化物的含量和谷胱甘肽过氧化物酶4的活性有关。化疗、放疗、分子靶向治疗、免疫治疗、中药抗肿瘤药物和纳米抗肿瘤药物都以不同的方式诱导肿瘤细胞发生铁死亡。以铁死亡作为靶点的抗肿瘤治疗有助于克服肿瘤细胞对现有治疗方法的耐受,并可恢复对治疗药物的敏感性。结论铁死亡是有前景的抗肿瘤治疗靶点,深入研究其在肿瘤中的作用机制并将其应用于临床抗肿瘤治疗,有望为肿瘤患者提供新的治疗方案。OBJECTIVE Ferroptosis,a new type of cell death different from autophagy,apoptosis and necrosis,is closely related to various human diseases.Tumor incidence increases with each passing year while therapeutic effect is not optimistic,so it is urgent to explore novel targets in tumor therapy.This study summaried the effect mechanisms of ferroptosis in tumor and advances in research on tumor therapy potentially targeted to ferroptosis including chemotherapy,radiotherapy,molecularly targeted therapy,immunotherapy,traditional Chinese medicine treatment and nanotechnology treatment.METHODS " Ferroptosis,chemotherapy,radiotherapy,molecularly targeted therapy and immunotherapy" were searched as keywords by PubMed and CNKI series full text database retrieval system from 2012-01 to 2020-01.Totally 120 English papers and 5 Chinese papers were obtained.Inclusion criteria:(1)discovery of ferroptosis;(2)functional mechanism of ferroptosis in tumor;(3)tumor therapy potentially targeted to ferroptosis.Exclusion criteria:(1)literature review;(2)lack of research data.According to the inclusion criteria and the exclusion criteria,50 papers were finally included in the analysis.RESULTS It is currently considered that the functional mechanism of ferroptosis is related to intracellular iron metabolism level,lipid peroxide content and glutathione peroxidase 4 activity.Chemotherapy,radiotherapy,molecularly targeted therapy,immunotherapy,traditional Chinese medicine treatment and nanotechnology treatment can result in tumor ferroptosis in different ways.Tumor therapies targeted to ferroptosis contribute to overcoming the resistance of tumor cells to existing therapies and can restore the sensitivity of tumor cells to therapeutic drugs.CONCLUSION The functional mechanism of Ferroptosis in tumor,apromising anti-tumor therapeutic target should be further studied,and it will bring new treatments to tumor patients if applied to clinical tumor therapy.
