摘 要:为优化东北刺人参不定根的总黄酮提取工艺,该试验在不同的提取溶剂、浓度、时间、温度、次数和料液比条件下对东北刺人参不定根中总黄酮含量进行筛选,得到最适宜的提取条件。结果表明:利用50%的乙醇进行提取,在提取时间2 h、提取温度50℃、提取次数2次、料液比1∶30(g∶mL)时的总黄酮含量最高,达到185.74 mg/g。同时,在此条件下该试验进一步测定了东北刺人参不定根提取物的抗氧化活性,结果表明,东北刺人参不定根提取物对DPPH和ABTS均有清除能力,在浓度为10 mg/mL时,ABTS自由基清除率达到了98.33%。而随着提取物浓度的升高,铁离子螯和能力逐渐增强,当达到一定浓度后,铁离子螯和能力保持平稳。To increase the content of total flavonoid in adventitious roots of Oplopanaxs elatus Nakai,an optimal extraction method was selected via determining the effects of solvents,extraction time,extraction temperature,extraction times,and ratio of material to liquid on flavonoid content.The results showed that the optimum extraction conditions were as follows:It's extracted with 50%ethanol,extraction time 2 h,extraction temperature 50℃,extraction times 2 times,ratio of material to liquid 1∶30;the total content was 185.74 mg/g.At the same time,the antioxidant activity of the extraction solvent was further evaluated under the optimum extraction conditions.The results showed that the adventitious root extract of ginseng under optimal conditions had the ability to clear both DPPH and ABTS.At the concentration of 10 mg/mL,the removal rate of ABTS free radicals reached 98.33%.However,with the increase of extract concentrations,the ability of iron ion chelation was positively correlated,and when a certain concentration was reached,the ability of iron ion chelation reached stable.
