雌激素对肺腺癌SPC-A-1细胞上皮间质转化标志物Vimentin、E-cadherin表达量的影响

摘　　要：[目的]探讨雌激素对肺腺癌SPC-A-1细胞系上皮间质转化(EMT)标志物E钙黏蛋白(E cadher-in).波形蛋白(Vimentin)表达量的影响。[方法]用不同浓度的雌激素刺激SPCA-1细胞系,并用qRT-PCR和Western-blot实验检测各组细胞中EMT标志物表达量的变化,用Transwell实验检测不同浓度雌激素对细胞迁移能力的影响,计算各组间有无统计学差异。[结果]当雌激素浓度为1×10-9 mol/L、1×10-8mol/L、1×10-7 mol/L时,Vimentin的mRNA表达量分别为(2.21±0.53).(4.81±0.66)、(2.27±0.51),显著高于空白对照组,组间有统计学差异(P<0.05),E-cadherin的mRNA表达量分别为(0.67±0.18)、(0.42±0.14)、(0.61±0.11),显著低于空白对照组(P<0.05),蛋白表达量也有同样趋势。细胞迁移数分别为(23.65±3.25)、(37.88土5.66)、(22.17±2.98),组间有统计学差异(P<0.05)。当雌激素浓度为1×10-8 mol/L时,Vimentin的表达量最高,E cadherin的表达量最低,细胞迁移数最高。[结论]适宜浓度雌激素可以促进Vimentin的表达,抑制E-cadherin的表达,提高细胞迁移能力,当雌激素浓度为1×10-8 mol/L时,促进Vimentin表达、抑制E-cadherin表达和促进细胞迁移的作用最显著,雌激素对SPC-A-1细胞系发生EMT过程有促进作用。[Objective]To investigate the effect of estrogen on the expression of epithelial mesenchymal transition markers of E-cadherin and vimentin in SPC-A-1 cell line.[Methods]SPC-A-1 cell line was stimulated with different concentrations of estrogen;the expresvsion of EMT markers was detected by Q-RT-PCR and Western blot;the effect of different concentrations of estrogen on cell migration was detected by Transwell test.The difference between the groups was calculated.【Results】When the estrogen concentration was 1×10-9mol/L、1×10-8 mol/L、1×10-1 mol/L,the Vimentin mRNA expression was(2.21±0.53),(4.81±0.66),(2.27±0.51),which was significantly higher than that of the blank control group(P<0.05).The mRNA expression of E-cadherin was(0.67d=0.18),(0.42±0.14),(0.61±0.11),which was significantly lower than that of the blank control group(P<0.05).The number of cell migration was(23.65±3.25),(37.88±5.66)and(22.17±2.98)respectively(P<0.05).When the estrogen concentration was 1×10-8mol/L»the expression of vimentin was the highest*the expression of E-cadhcrin was the lowest,and the number of cell migration was the highest.【Conclusion】Appropriate concentration of estrogen can promote the expression of vimentin.inhibit the expression of E-cadherin,and improve the ability of cell migration.When the concentration of estrogen is 1×10-8 mol/L,the effect of promoting vimentin expression,inhibiting E-cadherin expression and promoting cell migration is most significant.Estrogen can promote the EMT process of SPC-A-1 cell line.
