摘 要:目的探讨miRNA21联合重组人干扰素γ(IFN-γ)对小鼠肺癌移植瘤的作用效果,并分析了相关机制。方法每只小鼠接种2×10^6个肺癌细胞建立小鼠肺癌移植瘤模型。将建模成功的54只肺癌移植瘤小鼠随机分为3组:模型组、干扰素组与联合组,每组18只。模型组给予腹腔注射磷酸盐缓冲液治疗,干扰素组给予腹腔注射重组人IFN-γ治疗,联合组给予miRNA21联合重组人IFN-γ治疗。记录与计算小鼠的移植瘤重量和抑瘤率。采用TUNEL法检测与计算细胞凋亡指数。采用免疫组织化学法检测组织微血管密度(MVD)。采用Western bolt法检测Beclin-1、LC3的表达情况。采用免疫组化法检测肿瘤坏死因子-α(TNF-α)与白介素-6(IL-6)含量。结果①干扰素组与联合组的移植瘤重量[(4.91±0.15)、(4.01±0.22)g]低于模型组[(6.26±0.53)g],抑瘤率[(23.71±5.22)%、(40.86±11.31)%]高于模型组(0),且联合组的移植瘤重量低于干扰素组,抑瘤率高于干扰素组,差异有统计学意义(P<0.05)。②干扰素组与联合组的移植瘤细胞凋亡指数[(21.74±3.13)%、(30.67±4.14)%]均高于模型组[(3.13±0.24)%],且联合组移植瘤高于干扰素组,差异有统计学意义(P<0.05)。③干扰素组与联合组的移植瘤组织MVD计数(10.38±1.09、6.83±0.85)低于模型组(14.56±1.11),且联合组低于干扰素组,差异有统计学意义(P<0.05)。④干扰素组与联合组的移植瘤组织Beclin-1(6.34±0.34、16.43±1.91)、LC(37.23±0.82、12.22±1.13)相对表达量均高于模型组(1.13±0.03、1.23±0.11),且联合组移植瘤高于干扰素组,差异有统计学意义(P<0.05)。⑤干扰素组与联合组的血清TNF-α[(14.49±1.48)、(7.21±0.52)pg/mL]与IL-6含量[(16.02±1.85)、(8.08±1.84)pg/mL]均低于模型组[(27.33±1.49)、(34.20±2.11)pg/mL],且联合组低于干扰素组,差异均有统计学意义(P<0.05)。结论miRNA2与重组IFN-γ联合应用于小鼠肺癌移植瘤能抑制TNF-α与IL-6的表达,促进移植瘤细�Objective To explore the effects of miRNA21 combined with recombinant human interferonγ(IFN-γ)on lung cancer xenografts in mice,and analysis the related mechanisms.Methods Each mouse was inoculated with 2×10^6 lung cancer cells to establish a mouse lung cancer transplantation tumor model.54 cases of lung cancer xenograft tumor models successfully modeled were randomly divided into 3 groups-model group,interferon group and combined group,with 18 mice in each group.The model group were given intraperitoneal injection of phosphate buffer solution,the interferon group were given intraperitoneal injection of recombinant human IFN-γ,and the combination group were given miRNA21 combined with recombinant human IFN-γ.The tumor weight and tumor inhibition rate of the mice were recorded and calculated.Apoptosis index was detected by TUNEL method.Tissue microvessel density(MVD)was tested using immunohistochemistry.The expression of Beclin-1 and LC3 was detected by Western bolt method.The levels of tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)were detected by immunohistochemistry.Results①The weight of transplanted tumor in the interferon group and the combination group[(4.91±0.15),(4.01±0.22)g]was lower than that of the model group[(6.26±0.53)g],and the tumor inhibition rate[(23.71±5.22)%,(40.86±11.31)%]was higher than the model group(0),and the weight of the transplanted tumor in the combined group was lower than that of the interferon group,and the tumor inhibition rate was higher than that of the interferon group.The difference was statistically significant(P<0.05).②The xenograft cell index[(21.74±3.13)%,(30.67±4.14)%]of the interferon group and the combination group were higher than the model group[(3.13±0.24)%],and the combination group was higher than the interferon group,the difference was statistically significant(P<0.05).③The MVD counts of transplanted tumor tissue in the interferon group and the combination group(10.38±1.09,6.83±0.85)was lower than the model group(14.56±1.11),and the
