摘  要:以不同质量比的聚乳酸(PLA)和环丙沙星(CIP)作为靶材(PLA与CIP的质量比为1∶2、1∶1和2∶1),通过低功率电子束蒸发法(EBD)沉积3种比例的PLA基环丙沙星复合薄膜。采用X线光电子能谱仪(XPS)、核磁共振波谱仪(NMR)、场发射扫描电子显微镜(FESEM)和原子力显微镜(AFM)分析复合薄膜成分、结构和表面形貌,采用紫外可见分光光度计(UVVis)分析复合薄膜的药物释放动力学性能,运用琼脂扩散法测试复合薄膜抗菌性能及缓释抗菌性能。结果表明:3种PLA和CIP不同质量比的复合薄膜均可通过电子束蒸发法成功制备,复合薄膜在基底上分布较为均匀,呈现颗粒状或椭球状形貌,但薄膜样品的均方根粗糙度(Rq)相差明显,随着PLA含量的增加,Rq呈减小趋势;通过考查薄膜形貌结构、抗菌性能及动力学释放行为这几方面因素,得到靶材m(PLA)∶m(CIP)=1∶1时,复合薄膜的综合性能最佳,该比例的复合薄膜即使在生理盐水中浸泡168 h后,仍对金黄色葡萄球菌具有抗菌活性,与浸泡24 h后的复合膜相比,抑菌圈直径减少不超过37%。Polylactic acid(PLA)-based ciprofloxacin(CIP)composite films with different mass ratio(m(PLA)∶m(CIP)=1∶2,1∶1 and 2∶1)were prepared by low-energy electron beam deposition(EBD).The molecular structure,chemical state,morphology and kinetic release behavior of PLA-based ciprofloxacin composite films were investigated by X-ray photoelectron spectroscopy(XPS),nuclear magnetic resonance spectroscopy(NMR),field emission scanning electron microscope(FESEM),atomic force microscope(AFM)and ultraviolet and visible spectrophotomety(UV-Vis),the antibacterial property and sustained-release antibacterial property of composite films were tested by using the agar diffusion method.Results showed that the three kinds of composite films with different mass ratios could be successfully prepared by EBD.The composite films were uniformly distributed on the substrate,showing granular or spherical morphology,but the root mean square roughness(Rq)of the composite films was obviously different,and Rq tended to decrease with the increase of PLA.By investigating the morphology,antibacterial properties and kinetic release behavior of the composite films,the comprehensive properties of the composite films were the best when the mass ratio of targets m(PLA)∶m(CIP)was 1∶1.Even after soaking in saline for 168 h,the composite films still had antibacterial activity against S.aureus,and the diameter of the inhibition ring was reduced by no more than 37%compared with the composite films after being immersed for 24 h.
