摘 要:目的为探索临床常用药丹参多酚酸治疗脑梗死潜在的分子机制。方法采用网络药理学中药物成分的靶点预测、疾病相关靶点的数据挖掘、通路与功能的富集、可视化网络构建、分子对接等方法,分析了丹参多酚酸治疗脑梗死的蛋白靶点、相关通路及生物过程、成分与靶点之间结合能力。结果得到38个靶点,这些靶点参与调节免疫反应、活性氧族的代谢、抗氧化应激、调节凝血功能等生物过程。丹参多酚酸作用于过氧化物酶体增生物激活受体(PPAR)、白介素17(IL-17)、补体与凝血瀑布三条KEGG通路中的凝血酶原等13个靶点,干预脑水肿的形成、炎症反应、细胞凋亡、血栓形成及补体系统的激活等过程,达到神经保护作用。结论研究揭示了丹参多酚酸治疗脑梗死潜在的靶点、功能模块、通路及生物过程,为进一步研究提供了理论依据和参考。Objective To explore the potential molecular mechanism of salvianolic acids,which were commonly used in clinical practice for treating ischemic stroke.Method Network pharmacological methods were used to predict components’targets,mine related targets within databases,enrich targets’pathway and biological process,construct visual networks and dock molecular and protein.The molecular targets,signing pathway and biological process,binding ability between components and targets of salvia miltiorrhiza(SA)for treating brain ischemia were analyzed.Results Thirty-eight targets were obtained.These targets involved in biological processes like immune response,metabolism of reactive oxygen species,antioxidant stress,regulation of coagulation function,impacting PPAR,IL-17,complement and coagulation cascades KEGG pathway.SA intervenes the process of formation of cerebral edema,inflammatory response,cell apoptosis,thrombosis and activation of the complement system.In this way,SA generates neuroprotective effects.Conclusion This research uncovers the potential molecular targets,functional modules,signing pathways and biological processes of salvianolic acids for treating ischemic stroke,providing the basis and reference for next research.
