莱菔子抑制结肠癌干细胞恶性行为的研究

摘　　要：目的研究中药莱菔子对结肠癌干细胞恶性行为的影响,并探讨其作用机理。方法采用无血清悬浮培养法从HT29结肠癌细胞系中诱导培养结肠癌干细胞。制备莱菔子乙醇提取物,以不同浓度(30~120μg/mL)作用于结肠癌干细胞。采用悬浮成球法检测结肠癌干细胞的自我更新能力,采用CCK-8法检测增殖能力,采用Matrigel穿膜法检测侵袭能力,采用实时荧光定量PCR法检测结肠癌干细胞内长链非编码RNA HOTAIR的表达水平。结果莱菔子乙醇提取物有力抑制了结肠癌干细胞的自我更新能力、增殖能力和侵袭能力,显著下调了结肠癌干细胞内HOTAIR的表达水平。结论中药莱菔子能够抑制结肠癌干细胞的恶性行为,其作用机制与下调促癌的长链非编码RNA表达有关,在结肠癌的治疗中具有良好应用价值。Objective To study the effect of Laifuzi(Semen Raphani)on malignant behaviors of colon cancer stem cells andexplore the underlying mechanism.Methods Colon cancer stem cells were induced from HT29 colon cancer cell line by serum-free suspension culture.The ethanol extract of Laifuzi(Semen Raphani)were prepared and used to treat colon cancer stem cells at different concentrations(30~120μg/mL).The self-renewal ability of colon cancer stem cells was detected by suspension spheroidization assay,proliferation ability by CCK-8 assay,invasion ability by Matrigel assay,and expression level of long non-coding RNA HOTAIR in colon cancer stem cells by real-time fluorescence quantitative PCR.Results The ethanol extract of Laifuzi(Semen Raphani)inhibited the self-renewal,proliferation and invasion abilities of colon cancer stem cells,and significantly decreased the expression of HOTAIR in colon cancer stem cells.Conclusion Laifuzi(Semen Raphani)can inhibit the malignant behaviors of colon cancer stem cells and the mechanism is related to down-regulation of oncogenic long non-coding RNA expression,suggesting a good application value of Laifuzi(Semen Raphani)in the treatment of colon cancer.
