摘  要:通过在液体培养基中添加100μmol·L-1茉莉酸甲酯培养金针菇(Flammulina filiformis)菌丝,利用Label-free非标记定量蛋白质组学技术研究茉莉酸甲酯对金针菇菌丝体次级代谢的影响,并测定了代谢产物的含量。结果显示:与对照组比较,267个蛋白质的表达量上调,156个蛋白质的表达量下调,共有423个差异表达蛋白质;表达差异大于2倍的蛋白质中,32个蛋白质之间存在相互作用。KEGG富集分析显示,差异表达蛋白质主要富集于次级代谢途径,包括次级代谢产物、甾醇类化合物生物合成途径;其中,苯丙氨酸解氨酶和细胞色素P450单加氧酶分别上调3.92、7.54倍,呈极显著差异。三个萜烯类物质(齐墩果酸、熊果酸和山楂酸)的含量比对照组显著提高;激素类物质中,茉莉酸、茉莉酸异亮氨酸复合物、茉莉酸甲酯的含量分别是对照的18.84、33.53、3.18倍。研究结果为进一步探究茉莉酸甲酯对金针菇次级代谢的调控机制和信号转导提供参考。Label-free quantitative proteomics was used to analyze the effect of methyl jasmonate(MeJA,100μmol·L-1)on the secondary metabolism in Flammulina filiformis mycelium.Contents of secondary metabolites were also determined.There were 423 differentially expressed proteins in MeJA samples compared with the control(CK),of which 267 were up-regulated and 156 were down-regulated.Among proteins with a difference greater than 2 folds,32 proteins had protein-protein interactions.KEGG enrichment analysis showed that the differential proteins were mainly enriched in secondary metabolic pathways,including biosynthesis of secondary metabolites and steroid biosynthesis.Compared with the CK,phenylalanine ammonia lyase and cytochrome P450 monooxygenase were significantly up-regulated by 3.92 and 7.54 folds in these pathways,respectively.MeJA stimulated biosynthesis of oleanolic acid,ursolic acid and maslinic acid.Contents of jasmonic acid,jasmonic acid-isoleucine and methyl jasmonate in MeJA treated mycelia were 18.84,33.53 and 3.18 folds of those in the control group,respectively.These results provide a theoretical basis for further exploring the regulatory mechanism and signal transduction of methyl jasmonate on secondary metabolites in F.filiformis.
