摘 要:目的探讨乳腺癌患者miR-196a2基因rs11614913位点的多态性及其与癌基因表达变化的相关性。方法回顾性选择新疆医科大学附属肿瘤医院2017年3月至2019年12月期间进行乳腺癌根治术的64例患者作为乳腺癌组,同期在新疆医科大学附属肿瘤医院体检的80例健康志愿者作为对照组。测定2组受试者外周血中miR-196a2基因rs11614913位点的多态性以及乳腺癌病灶内抑癌基因[程序性细胞死亡因子4(PDCD4)、细胞周期蛋白5(CCN5)、伴有kazal基序富含半胱氨酸的逆转诱导蛋白(RECK)、转录延伸因子A样蛋白7(TCEAL7)]、原癌基因[Livin、生存素(Survivin)、细胞周期蛋白D1(Cyclin D1)、基质金属蛋白酶(MMP2)、N-钙黏蛋白(N-cadherin)]的mRNA表达量。结果乳腺癌组患者miR-196a2基因rs11614913位点C/C基因型的分布频率高于对照组(χ2=11.827,P<0.05),T/T基因型的分布频率低于对照组,差异有统计学意义(χ2=10.474,P<0.05),C/T基因型的分布频率与对照组比较,差异无统计学意义(χ2=0.002,P>0.05);miR-196a2基因rs11614913位点C/C基因型乳腺癌病灶内PDCD4、CCN5、RECK、TCEAL7的mRNA表达量明显低于T/T基因型及T/C基因型的乳腺癌病灶,Livin、Survivin、Cyclin D1、MMP2、Ncadherin的mRNA表达量明显高于T/T基因型及T/C基因型的乳腺癌病灶,差异有统计学意义(P<0.05)。结论乳腺癌患者存在miR-196a2基因rs11614913位点等位基因T向C的突变并且能够影响原癌基因、抑癌基因的表达。Objective To investigate the polymorphism of miR-196 a2 gene rs11614913 locus and its correlation with oncogene expression in breast cancer patients.Methods Sixty-four patients who underwent radical mastectomy in the Affiliated Tumor Hospital of Xinjiang Medical University during March 2017 to December 2019 were selected retrospectively as the breast cancer group and 80 healthy volunteers who underwent physical examination in the same period were selected as the control group.The polymorphisms of microRNA196 a2 gene at rs11614913 locus in peripheral blood and the mRNA expression of tumor suppressor gene[programmed cell death factor 4(PDCD4),Cyclin 5(CCN5),reversion-inducing cysteine-rich protein with Kazal motif(RECK),transcription elongation factor A-like protein 7(TCEAL7)],proto-onco gene[Livin,Survivin,Cyclin D1,matrix metalloproteinase(MMP2),N-cadherin]in breast cancer lesions were measured.Results The composition ratio of C/C genotype at rs11614913 locus of miR-196 a2 gene in breast cancer group was higher than that in control group(χ2=11.827,P<0.05),the composition ratio of T/T genotype was lower than that in control group(χ2=10.474,P<0.05),and the composition ratio of C/T genotype had no significant difference with that in control group(χ2=0.002,P>0.05).The mRNA expression of PDCD4,CCN5,RECK,TCEAL7 in breast cancer with C/C genotype were significantly lower than those in breast cancer with T/T genotype and T/C genotype,mRNA expression of Livin,Survivin,Cyclin D1,MMP2,N-cadherin were significantly higher than those in breast cancer with T/T genotype and T/C genotype.Conclusion MiR-196 a2 gene in breast cancer patients exists mutation of allele T to C at rs11614913 locus and can affect the expression of proto-oncogene and tumor suppressor gene.
