摘  要:目的研究KCC2在大鼠脊髓过表达后对大鼠吗啡耐受中NMDAR和PKC表达的的影响。方法构建KCC2腺病毒表达载体,将30只SD雄性大鼠随机分为三组,Ⅰ组为KCC2过表达组,Ⅱ组为GFP对照组,Ⅲ组为空白对照组,每组大鼠为10只。Ⅰ组大鼠鞘内注射病毒上清液(108/ml)20μL,Ⅱ组鞘内注射control vector 20μL,Ⅲ组鞘内注射生理盐水20μL。2周后,检测KCC2过表达,各组通过脊髓置管注入吗啡,每日2次。吗啡注射量为15 ug/kg,连续7 d,检测KCC2表达,检测NMDAR和PKC表达。结果与对照组和空白组相比,KCC2过表达抑制吗啡耐受中NMDAR的表达,PKC表达降低。结论KCC2脊髓过表达模型大鼠吗啡耐受过程中,NMDAR和PKC表达降低。Objective To investigate the effect of KCC2 overexpression in rat spinal cord on the expression of NMDAR and PKC in morphine tolerance.Methods KCC2 adenovirus expression vector was constructed,30 male SD rats were randomly divided into three groups,Ⅰfor KCC2 gene therapy group,Ⅱgroup for GFP,Ⅲgroups as blank control group,each group of rats to 10.Ⅰgroup rats intrathecal injection on virus supernatant(108/ml)20μL,Ⅱintrathecal injection control group vector20μL,Ⅲgroup of intrathecal injection of saline 20μL.After 2 weeks,KCC2 overexpression was detected,and morphine was injected into each group through spinal cord catheterization,twice a day.The expression of KCC2 and NMDAR and PKC were detected after morphine injection of 15ug/kg for 7 consecutive days.Results Compared with control group and blank group,KCC2 overexpression inhibited NMDAR expression and PKC expression in morphine tolerance.Conclusion NMDAR and PKC expression were decreased during morphine tolerance in rats with KCC2 spinal cord overexpression model.
