摘  要:目的探讨黄连素对人结肠癌细胞株THC-8307奥沙利铂(OXA)耐药性的影响。方法(1)人结肠癌细胞株THC-8307和耐OXA结肠癌细胞株THC-8307/OXA均分为空白对照组、黄连素5μg/mL组、黄连素10μg/mL组、黄连素20μg/mL组进行实验,除空白对照组(不加入任何药物)外,各组加入相应浓度的黄连素进行干预。干预48 h后,观察各组细胞形态并检测各组细胞存活率。(2)THC-8307细胞和THC-8307/OXA细胞均分为OXA对照组、黄连素5μg/mL+OXA组、黄连素10μg/mL+OXA组、黄连素20μg/mL+OXA组进行实验,给予相应的药物进行干预48 h后,观察各组细胞形态,检测各组细胞存活率,并计算各组的半数抑制浓度(IC 50)及逆转倍数。结果(1)空白对照组、黄连素5μg/mL组、黄连素10μg/mL组、黄连素20μg/mL组的THC-8307细胞和THC-8307/OXA细胞形态和生长状态无明显差异,各组细胞存活率比较差异均无统计学意义(均P>0.05)。(2)在THC-8307细胞中,OXA对照组、黄连素5μg/mL+OXA组、黄连素10μg/mL+OXA组、黄连素20μg/mL+OXA组的细胞形态、生长状态、存活率均无差异。在THC-8307/OXA细胞中,OXA对照组、黄连素5μg/mL+OXA组、黄连素10μg/mL+OXA组、黄连素20μg/mL+OXA组的细胞存活率依次降低,各黄连素组的IC 50低于OXA对照组,而逆转倍数高于OXA对照组(均P<0.05)。结论黄连素可逆转耐OXA结肠癌细胞株THC-8307/OXA的耐药性,从而提高OXA对细胞的抑制作用。Objective To explore the effect of berberine on oxaliplatin(OXA)resistance in human colon cancer cell line THC-8307.Methods(1)Both human colon cancer cell line THC-8307 and OXA-resistant colon cancer cell line THC-8307/OXA were divided into blank control group,5μg/mL berberine group,10μg/mL berberine group,and 20μg/mL berberine group in the experiment,except the blank control group(not given any drugs),all groups were added with corresponding concentrations of berberine for intervention.After 48 hours of intervention,cell morphology was observed and cell survival rate was detected in each group.(2)Both THC-8307 cells and THC-8307/OXA cells were divided into OXA control group,5μg/mL berberine+OXA group,10μg/mL berberine+OXA group and 20μg/mL berberine+OXA group in the experiment.After 48 hours of intervention with corresponding drugs,cell morphology was observed and cell survival rate was detected in each group,and median inhibition concentration(IC 50)and reversal fold were calculated in each group.Results(1)No significant differences were found in morphology or growth status of THC-8307 cells/THC-8307/OXA cells among the blank control group,the 5μg/mL berberine group,the 10μg/mL berberine group and the 20μg/mL berberine group,cell survival rate showed no statistically significant difference among groups(all P>0.05).(2)In terms of THC-8307 cells,there was no difference in cell morphology,growth status or survival rate among the OXA control group,the 5μg/mL berberine+OXA group,the 10μg/mL berberine+OXA group and the 20μg/mL berberine+OXA group.In terms of THC-8307/OXA cells,cell survival rates of the OXA control group,the 5μg/mL berberine+OXA group,the 10μg/mL berberine+OXA group and the 20μg/mL berberine+OXA group decreased in turn,the berberine goups had lower IC 50 and higher reversal fold than the OXA control group(all P<0.05).Conclusion Berberine can reverse the drug resistance of OXA-resistant colon cancer cell line THC-8307/OXA,thus improving the inhibitory effect of OXA on the cells.
