摘 要:探讨益生菌鼠李糖杆菌GG(LGG)对小鼠急性肝损伤的保护作用的影响.本研究以C57BL/6小鼠为实验对象,分为对照组、酒精组和酒精+LGG组,用LGG或MRS培养基将小鼠连续处理4周后再采用50%(V/V)的乙醇灌胃,建立小鼠急性肝损伤模型,对各组小鼠外周血中ALT和AST的含量及肝脏TG含量进行检测,观察肝脏组织病理学变化及肝脏脂质代谢相关基因的表达.结果发现,与对照组相比,酒精组小鼠外周血中ALT和AST的含量及肝脏TG含量显著升高,肝脏组织脂肪合成相关基因表达显著升高,脂肪分解相关基因表达显著降低;与酒精组相比,酒精+LGG组的以上指标均呈现不同程度的逆转.表明LGG具有保护小鼠急性酒精性肝损伤的作用,并且其保护作用是通过调节肝脏脂质代谢实现的.This paper investigates the protective effect of the probiotic Lactobacillus rhamnosus GG(LGG)against acute liver injury in mice.In the experiment,C57BL/6 mice were selected as the experimental subjects,which were divided into the control group,the alcohol group,and the alcohol+LGG group.After the LGG or MRS culture medium was continuously applied to process the mice for 4 weeks,the mice were given intragastric administration with 50%(V/V)ethanol,thus establishing a model of acute liver injury of mice.The contents of serum ALT and AST and liver TG were tested to observe liver tissue pathological changes and the expression of liver lipid metabolism-related genes.Results show that compared with the control group,the alcohol group has significantly higher levels of serum ALT and AST and liver TG,a significantly increased expression of genes related to lipid synthesis in liver tissues,and a significantly decreased expression of genes related to lipolysis.Meanwhile,compared with the alcohol group,the above indicators in the alcohol+LGG group are reversed to varying degrees.All those indicate that LGG can protect mice from acute alcoholic liver injury,and its protective effect is realized through regulating liver lipid metabolism.
