摘  要:目的比较牙龈成纤维细胞在聚醚醚酮(polyetheretherketone,PEEK)以及纯钛(titanium,Ti)表面的生物相容性。方法制备直径为10.0 mm,厚度为1.0 mm的圆形片状PEEK试件和Ti试件。将PEEK试件和Ti试件分别与人牙龈成纤维细胞细胞在体外共培养,采用扫描电镜观察试件表面形貌以及细胞黏附形态、CCK-8检测细胞活性与增殖能力、calcecin-AM/PI双染细胞方法检测活细胞和死细胞、鬼笔环肽染色检测细胞形态。使用免疫荧光,CCK-8等方法对比人牙龈成纤维细胞细胞在PEEK试件和Ti试件表面的细胞黏附、增殖及形态改变。结果(1)人牙龈成纤维细胞在PEEK试件和Ti试件表面培养24 h后细胞在PEEK表面黏附优于Ti表面;(2)人牙龈成纤维细胞细胞接种于PEEK试件和Ti试件表面培养的6 h、12 h、24 h,PEEK组的活细胞数均高Ti组;(3)人牙龈成纤维细胞细胞与试件共培养的6 h、12 h、24 h,PEEK组的细胞增殖能力高于Ti组(P<0.05);(4)鬼笔环肽染色观察细胞形态结果显示,牙龈成纤维细胞在PEEK表面黏附能力优于Ti表面。结论PEEK体外生物相容性良好,且优于Ti组,PEEK能够促进牙龈成纤维细胞的黏附、增殖,有利于软组织黏附,利于种植体美学修复。Objective To compare the biocompatibility of gingival fibroblasts on polyetheretherketone(PEEK)and pure titanium(Ti)surfaces.Methods Preparing a round piece of PEEK and Ti specimen with diameter of 10.0 mm and thickness of 1.0 mm.The PEEK and Ti specimen were co-cultured with human gingival fibroblast cells in vitro,and the surface morphology and cell adhesion morphology were observed by scanning electron microscopy(SEM).CCK-8 method was used to detect cell activity and proliferation,t calcecin-AM/PI Double staining cell method to detect live and dead cells,and the cell morphology was detected by phlebiopeptide staining.Immunofluorescence and CCK-8 were used to compare the cell adhesion,proliferation and morphological changes of human gingival fibroblasts on the surfaces of PEEK and Ti specimens.Results(1)Human gingival fibroblasts were cultured on the surface of PEEK specimens and Ti specimens for 24 h,the cells adhered better to the surface of PEEK than Ti surfaces;(2)Human gingival fibroblast cells were seeded on PEEK and Ti specimens after cultured for 6 h,12 h and 24 h,the number of viable cells in the PEEK group was higher than that in the Ti group;(3)The cell proliferation ability of the PEEK group was higher than Ti group after co-cultured with human gingival fibroblast cells for 6 h,12 h and 24 h(P<0.05);(4)The cell morphology observed by phalloidin staining showed that the adhesion ability of gingival fibroblasts on PEEK surface was better than Ti surface.Conclusion PEEK has good biocompatibility in vitro and is superior to Ti group.PEEK can promote the adhesion and proliferation of human gingival fibroblasts,which is beneficial to soft tissue adhesion and implant aesthetic repair.
