摘 要:本试验探讨了阿魏酸(FA)与连翘苷(PHI)联合对LPS诱导奶牛乳腺上皮细胞(BMEC)炎性损伤的保护作用,为中药防治大肠杆菌性奶牛乳房炎提供依据。将BMEC分为空白对照组(Con)、模型组(LPS)、FA组(FA15+LPS)、PHI组(PHI5+LPS)、低浓度联合用药组(FA5+PHI5+LPS)、高浓度联合用药组(FA15+PHI5+LPS)。FA、PHI预处理BMEC 2 h后,加入LPS诱导12 h,采用流式细胞术检测细胞内氧自由基(ROS)含量及细胞凋亡率;RT-PCR法检测细胞内IL-1β、IL-6、TNF-α的表达量;试剂盒检测细胞内丙二醛(MDA)含量,超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)的活性。结果表明:LPS诱导BMEC胞内ROS、IL-1β、IL-6、TNF-α含量显著升高;SOD、GSH-Px活性显著降低、MDA含量显著升高;细胞凋亡率显著升高。低、高浓度联合用药组均显著抑制了LPS诱导的BMEC炎症、氧化应激指标变化,减少了细胞凋亡率,且效果优于FA或PHI单独用药组。研究表明,FA、PHI对LPS诱导的BMEC炎性损伤具有协同保护作用。This study investigated the protective effect of ferulic acid(FA)in combination with phillyrin(PHI)on LPS-induced inflammatory injury of dairy cow mammary epithelial cells(BMEC)in the hope of providing some basis for the treatment of coliform mastitis in dairy cows by clearing heat and detoxifying traditional Chinese medicine.Here,BMEC was divided into the control group(Con),the model group(LPS),the FA group(FA15+LPS),the PHI group(PHI5+LPS),the low concentration combination group(FA5+PHI5+LPS)and the high concentration combination group(FA15+PHI5+LPS).After FA and PHI pretreatment of BMEC for 2 h,LPS was added to induce for 12 h.The intracellular ROS content and apoptosis rate were detected by flow cytometry;Intracellular IL-1β,IL-6,and TNF-αwere detected by RT-PCR;and The content of malondialdehyde(MDA)andthe activity of superoxidedismutase(SOD)and glutathione peroxidase(GSH-Px)were detected by the kits.The results were that the levels of ROS,IL-1β,IL-6 and TNF-αwere significantly increased and the activities of SOD and GSH-Px were decreased;and the content of MDA was increased significantly and the apoptosis rate was notably increased by LPS in BMEC.Both the low-concentration combination group and the high-concentration combination group significantly inhibited the changes in LPS-induced inflammation and oxidative stress indicators,and reduced the rate of cell apoptosis.These results were better than those of the FA or PHI groups.In conclusion,FA and PHI have synergistic protective effects on LPS-induced BMEC ininjury caused by inflammation and oxidative stress.
