摘  要:目的通过构建结肠癌多药耐药细胞系为化疗效果早期预测和治疗策略的选择提供理论依据。方法首先通过浓度梯度法建立CRC多药耐药细胞系;其次采用CCK-8及Western实验检测细胞活性及耐药蛋白表达。结果成功建立CRC多药耐药细胞系,高浓度耐药细胞较低浓度耐药细胞具有更高耐药发生率,差异有统计学意义(P<0.05);较敏感细胞而言,2种耐药细胞株增殖能力无显著变化,差异无统计学意义(P>0.05);耐药细胞活力较敏感细胞增强,差异有统计学意义(P<0.05);Western实验检测较敏感细胞而言,耐药细胞P-gp蛋白表达增高,差异有统计学意义(P<0.05)。结论浓度递增法成功构建CRC多药耐药细胞系,为CRC耐药机制研究建立了平台,高浓度耐药细胞更有利于多药耐药的研究。Objective To establish a multi-drug resistance cell line of colorectal carcinoma(CRC)for early prediction and selection of treatment strategy.Methods CRC multidrug resistance cell lines were established by the concentration gradient method;and the cell activity and expression of drug resistance protein were detected by CCK-8 and Western blot.Results CRC multi-drug resistant cell lines were successfully established.The incidence of drug resistance was higher in the high concentration group than in the low concentration group(P<0.05).There was no significant change in the proliferative ability of the two drug-resistant cell line groups(P>0.05).The activity of drug resistant cells with high concentration was higher than that of the control group(P<0.05).The expression of P-gp protein in the drug-resistant cells with high concentration was significantly higher than that in the control group by western assay(P<0.05).Conclusion CRC multi-drug resistance cell line was successfully constructed by the increasing concentration method,which provides a platform for the studying mechanism of CRC drug resistance.
