摘  要:目的探讨乌梅总黄酮(FMF)对6-羟基多巴胺(6-OHDA)诱导的帕金森病大鼠脑线粒体呼吸链酶复合物的活性及相关线粒体ND1、ND4基因及蛋白表达的影响。方法采用6-OHDA左侧纹状体注射建立帕金森病大鼠模型。大鼠随机分成假手术组,模型组,FMF高、中、低剂量治疗组,每组各10例。FMF高、中、低剂量组予以FMF灌胃给药,剂量分别为160、80、40 mg·kg^-1,假手术组及模型组大鼠接受等容积生理盐水干预。每日1次,连续给药4周。采用比色法检测各组大鼠脑线粒体呼吸链酶复合物Ⅰ、Ⅱ、Ⅲ和Ⅳ的活性,采用RT-PCR检测编码线粒体呼吸链酶复合物Ⅰ的ND1、ND4 mRNA,采用Western Blot法检测ND1、ND4蛋白表达。结果与假手术组比较,模型组大鼠脑线粒体呼吸链酶复合物Ⅰ、Ⅱ、Ⅲ、Ⅳ活性明显降低(P<0.05);与模型组比较,FMF高剂量组大鼠脑线粒体呼吸链酶复合物Ⅰ~Ⅳ活性明显改善,FMF中剂量组线粒体呼吸链酶复合物Ⅰ、Ⅱ、Ⅳ的活性也有不同程度的提高(P<0.05),FMF低剂量组线粒体呼吸链酶复合物Ⅰ~Ⅳ活性无明显改变(P>0.05)。与假手术组比较,模型组ND1、ND4 mRNA及其蛋白的表达均显著下降(P<0.05);与模型组比较,FMF中、高剂量组均可提高ND1、ND4 mRNA及其蛋白的表达(P<0.05),其中以FMF高剂量组效果最为显著,而FMF低剂量组仅可提高ND4 mRNA的表达(P<0.05)。结论 FMF可能是通过影响线粒体呼吸链酶复合物的活性,提高线粒体ND1、ND4基因和蛋白的表达,防治帕金森病。Objective To investigate the effects of Fructus mume total flavone(FMF) on the activities of mitochondrial respiratory chain complexes and the expression of relative mitochondrial genes of ND1,ND4 mRNA in rats with Parkinson’s disease. Methods Model rats were established by striatum perfusion on the left side with 6-OHDA. The experimental rats were randomly divided to the sham operation group,the model group,and the FMF treatment groups with low,medium and high dose of FMF,as each group had 10 rats. FMF was given orally by gavage to FMF treatment group rats at doses of 160,80,40 mg·kg^-1 respectively,while the equal volume of normal saline was given to sham operation group and model group respectively. Four weeks after treatment,the enzyme activities of mitochondrial respiratory chain complexes in rats from each group were monitored by colorimetric method,RT-PCR and Western Blot were used to measure the expression of ND1,ND4 mRNA on mitochondrial of brain tissues of rats. Results As compared with the sham operation group,the activities of respiratory chain complexes I-Ⅳ were decreased significantly in model group(P<0.05). As compared with the model group,the activities of respiratory chain complexes Ⅰ-Ⅳ were increased significantly in high dose FMF group,activities of complexes Ⅰ,Ⅱ,Ⅳ were also increased obviously in medium dose FMF group(P<0.05). However,activities of complexes Ⅰ-Ⅳ showed no statistical significance in low dose FMF group(P>0.05). RT-PCR and Western Blot results showed that as compared with the sham group,the expression of ND1 and ND4 mRNA of rat brain tissues in the mitochondrial were significantly decreased in model group(P<0.05). As compared with the model group,the expression of ND1 and ND4 mRNA was increased significantly in high,middle dose FMF groups(P<0.05). The effect of high dose FMF was most obvious.
