摘  要:为研究牛等大型家畜肠道菌群的多样性及饲草性纤维素分解菌的背景,从东北及内蒙牛养殖主产区和屠宰加工区环境中采集大量动物毛发、粪便、肉样、血液样品,共计分离620株纤维化微小菌。对其中200株进行了纤维素酶等筛选,共79株具有较为明显的纤维素酶活性,其中1株从牛粪便中分离的纤维化微小菌(72-3)的初步分析结果表明,该菌纤维素酶活性较高,具有分泌纤维素酶、蛋白酶、甘露聚糖酶、葡聚糖酶、几丁质酶、果胶酶、木聚糖酶等多种酶的能力。经基质辅助激光解吸飞行时间质谱仪MALDI-TOF、法国梅里埃VITEK生化鉴定仪和16S rDNA鉴定,确定为放线菌门中的纤维化微小菌(Cellulosimicrobium sp.nov.),经小鼠急性毒性试验表明,该株菌具有低致病性,对动物是安全的。药敏试验结果显示,该菌对24种抗菌药中的20种耐药,仅对4种抗生素头孢哌酮、头孢曲松、磺胺异恶唑、阿奇霉素敏感。这一研究首次从家畜牛体内分离出纤维化微小菌,为家畜肠道内菌群多样性及纤维素消化等研究提供相关依据,也为后续纤维化微小菌的相关酶学性质开发利用奠定科学基础。In order to study the diversity of intestinal microflora of cattle,sheep and other livestock and the situation of forage cellulose decomposing bacteria,a large number of animal hair,feces and blood samples were collected from the environment of the main production area and slaughtering processing area of cattle and sheep in Northeast China,620 strains of fibrotic microflora were isolated in total.Among them,200 strains were screened for cellulase,79 strains had obvious cellulase activity,and one of them(72-3)as Cellulosimicrobium sp.was isolated from cattle faeces.The results showed that the cellulase activity of the strain was high,and it could secrete cellulase,protease,mannanase,glucanase,chitinase,pectinase,xylanase and other enzymes.The strain was identified as Cellulosicmicrobium sp.nov.by MALDI-TOF,VITEK and 16 S rDNA.The acute toxicity test of mice showed that the strain had low pathogenicity and was safe for animals.The results of drug sensitivity test showed that the strain was resistant to 20 of the 24 antibiotics,but sensitive to 4 antibiotics:cefoperazone,ceftriaxone,sulfamethoxazole and azithromycin.This study is the first time to isolate micro fibrobacteria from livestock and cattle,which provides relevant basis for the diversity of intestinal flora and cellulose digestion of livestock,and lays a foundation for the development and utilization of related enzyme properties of Cellulosimicrobium sp.nov.
