摘  要:SWEETs是一种广泛存在于真核生物中的糖转运蛋白,在植物的生长发育过程中具有重要作用。为研究马铃薯中SWEET11基因具体的生物学功能,本研究以马铃薯品种‘青薯9号’为试验材料,通过qRT-PCR技术测定StSWEET11在蔗糖、果糖、葡萄糖胁迫下的相对表达量。结果显示,在3%果糖处理下,StSWEET11的相对表达量在3 h、6 h、12 h和24 h均低于对照组,在48 h时恢复正常。在3%蔗糖处理下,StSWEET11基因呈先下降后上升的趋势,相对表达量均高于对照组。在3%葡萄糖处理下,StSWEET11基因的表达呈下降-上升-下降的趋势。通过测定转基因烟草和野生型烟草中的可溶性糖含量发现,转基因烟草中蔗糖含量高于野生型,果糖含量低于野生型,葡萄糖含量与野生型烟草无显著差异。此外,在蔗糖培养基上,转基因植株生长优于野生型植株,在葡萄糖培养基上,转基因植株和野生型植株生长无显著差异,而在果糖培养基上,转基因植株生长弱于野生型植株。结果表明,StSWEET11基因在马铃薯中对蔗糖的转运起正调控作用,对果糖的转运起负调控作用,不参与葡萄糖的转运。The SWEETs family is a sugar transporter protein found in eukaryotes and plays an important role in plant growth and development. In order to study the function of SWEET11 gene in potatoes, study took potato’Qingshu NO.9’ as the test material to determine the expression of StSWEET11 gene under the stress of different sugar by q RT-PCR. The results showed that under the treatment of 3% fructose, the relative expression level of StSWEET11 was lower than that of the control group at 3 h, 6 h, 12 h and 24 h, and returned to normal at 48 h. Under 3% sucrose treatment, StSWEET11 gene firstly showed a downward trend then increased, and its relative expression level was higher than that of the control group, StSWEET11 gene expression showed a down-up-down trend under 3% glucose treatment. By measuring the content of soluble sugar in transgenic tobacco and wild tobacco, it was found that the content of sucrose and fructose in transgenic tobacco was higher than that in wild tobacco,and the content of glucose was significantly different from that in wild-type tobacco. Phenotypic observation showed that transgenic plants grew better than wild-type plants on sucrose medium, and there was no significant difference between transgenic plants and wild plants on glucose medium, while transgenic plants on fructose medium grew less than wild plants. Therefore, it is speculated that StSWEET11 gene plays a positive regulatory role in the transport of sucrose and a negative regulatory role in the transport of fructose in potatoes, but has no transport function in glucose.
