摘  要:携带大片段基因组DNA的细菌人工染色体(bacterial artificial chromosome, BAC)文库是进行基因组结构与功能等相关研究的重要基因组资源。澳洲棉是二倍体野生棉,具有抗或耐棉蚜、抗黄萎病、抗干旱、增加衣分率、提高纤维成熟度的潜力,种子无腺体或植株有腺体等许多优良性状。本研究是在现有的野生棉二倍体和四倍体棉BAC文库基础上,以二倍体野生棉种澳洲棉为实验材料,提取澳洲棉幼嫩叶的DNA,经过HindⅢ酶切片段化,克隆到pSMART BAC (HindⅢ)载体,构建了澳洲棉的基因组细菌人工染色体(bacterial artificial chromosome, BAC)文库。该文库总共包含16 128个克隆(42个384孔板),平均插入的片段为120 kb,基因组覆盖率约1.06倍。澳洲棉BAC文库的构建将为棉属野生种质资源的保存与利用积累重要基因组资源。Bacterial artificial chromosome(BAC) library carrying large fragments of genomic DNA is an important genomic resource for genomic structure and function research. Gossypium australe is one kind of diploid wild cotton, and has many good traits such as resistance to cotton aphids, Verticillium wilt, drought, potential of increasing the percentage of lint and fiber maturity, and no glands in seeds or plants. In this study, based on the existing wild cotton diploid and tetraploid cotton BAC library, DNA of young leaves of diploid wild cotton species G.australe was extracted and fragmentated by Hind Ⅲ and cloned into pSMART BAC(Hind Ⅲ) vector to construct a bacterial artificial chromosome(BAC) library of G. australe. The library contains a total of 16 128 clones, and the average inserted fragment is 120 kb. Based on an estimated genome size of G. australe, the BAC library was estimated to have a total coverage of 1.06 ×genome equivalents. The construction of the G. australe BAC library will accumulate important genomic resources for the conservation and utilization of wild cotton germplasm resources.
