摘 要:鸟王茶是贵州省重要的经济作物之一,近年来其种植规模逐渐扩大。为了更好地研究鸟王茶优质品质的稳定遗传、大量增殖以及快速繁殖,本研究以鸟王茶NW32带腋芽茎段为外植体,研究外植体的灭菌条件、腋芽萌发与增殖诱导以及壮苗与生根的最佳培养基配方、炼苗及移栽的最佳处理条件。结果表明:(1) 75%酒精浸泡60 s,0.1%升汞浸泡13 min为最佳灭菌处理条件,该处理下污染率10.00%较低、成活率86.36%较高、并且死亡率1.21%较低。(2) MS+2.0 mg/L 6-BA+0.8 mg/L IBA+1.0 mg/L GA3为腋芽萌发最佳培养基,其诱导率高达95.56%。(3) MS+3.0 mg/L 6-BA+0.2 mg/L IBA+1.0 mg/L GA3为最佳增殖培养基,增值系数6.59为最高。(4) MS+2.0 mg/L 6-BA+0.8 mg/L IBA为最佳壮苗培养基,株高差为2.38 cm。(5) 50.0 mg/L IBA无菌溶液浸泡5 min,接种至1/2MS+1.5 mg/L IBA为最佳生根培养基,生根率高达100.00%。(6)以自然光照室温炼苗5 d,以V营养土∶V珍珠岩∶V蛭石=2∶1∶1为基质为最佳炼苗及移栽条件,其成活率高达90.00%。本研究获得了一套鸟王茶NW32完整的组培快繁体系,为鸟王茶工厂化育苗与规模化种植提供技术参考。Niaowang tea is one of the important economic crops in Guizhou province, and its planting scale has gradually expanded in recent years. In order to better stabilize its high quality, inherit, mass proliferation and rapid reproduction, in this study, the axillary bud stem of the NW32 of Niaowang tea were used as explants to study the sterilization conditions of the explants, the germination and proliferation induction of axillary buds, and the best medium formula, seedling and transplanting of the strong seedlings and rooting, processing conditions. The results showed that:(1) The best sterilization treatment condition is 75% alcohol soak for 60 s, 0.1% mercury immersion for 13 min, Under this treatment, the pollution rate was lower(10.00%), the survival rate was higher(86.36%), and the mortality rate was lower(1.21%).(2) The best medium for axillary bud germination is MS+2.0 mg/L 6-BA+0.8 mg/L IBA+1.0 mg/L GA3, the induction rate was as high as 95.56%.(3) The highest proliferation medium is MS+3.0 mg/L 6-BA+0.2 mg/L IBA+1.0 mg/L GA3, the proliferation coefficient of 6.59 is the highest.(4) The beststrong seedling medium is MS+2.0 mg/L 6-BA+0.8 mg/L IBA, and the plant height difference is 2.38 cm.(5) The best rooting medium is 50.0 mg/L IBA sterile solution for 5 min, inoculated to 1/2 MS+1.5 mg/L IBA,rooting rate is up to 100.00%.(6) The best conditions for refining and transplanting seedlings is the seedlings were incubated for 5 days at room temperaturewith natural light, and Vnutrient soil∶Vperlite∶V vermiculite=2∶1∶1 as the matrix, the survival rate was as high as 90.00%. In this study, a complete tissue culture and rapid propagation system of the NW32 of Niaowang tea was obtained, which Provide technical reference for the planting and large-scale planting of Niaowang tea.
