摘  要:目的:检测KCTD10在非小细胞肺癌中的表达水平及其对Hippo通路的影响。方法:采用免疫组化方法检测KCTD10在非小细胞肺癌组织标本中的表达;采用Western blot检测YAP、P-YAP、P-LATS1、MMP7、CyclinE在非小细胞肺癌细胞中的表达;采用集落形成实验和Transwell实验检测细胞增殖和迁移。结果:KCTD10在非小细胞肺癌组织及细胞系中高表达;KCTD10过表达促进A549细胞增殖和迁移;过表达KCTD10的A549细胞系YAP的表达总量未见明显变化,但抑制了P-YAP、P-LATS1的表达,促进了MMP7、CyclinE的表达。结论:KCTD10抑制了非小细胞肺癌中Hippo通路活性;KCTD10可能作为非小细胞肺癌防治的一个潜在靶向分子。Objective:To detect the expression level of KCTD10 in non-small cell lung cancer and its influence on the Hippo pathway.Methods:Immunohistochemical method was used to detect the expression of KCTD10 in non-small cell lung cancer tissue samples.Western blot was used to detect the expression of YAP,P-YAP,P-LATS1,MMP7,and CyclinE in non-small cell lung cancer cells.Colony formation and the Transwell assay were used to detect the proliferation and migration ability of cells.Results:KCTD10 was highly expressed in non-small cell lung cancer tissues and cell lines.KCTD10 overexpression promoted the proliferation and migration of A549 cells.The total expression of YAP in KCTD10-overexpressing A549 cell line did not change significantly,but it suppressed the expression of P-YAP and P-LATS1,and promoted the expression of MMP7 and CyclinE.Conclusion:KCTD10 inhibits the activity of Hippo pathway in non-small cell lung cancer.KCTD10 may be a potential targeting molecule for the prevention and treatment of non-small cell lung cancer.
